摘要
目的比较斑块状寻常型银屑病(psoriasis vulgaris,PV)和汗孔角化症(porokeratosis,PK)患者的外周血Vγ9Vδ2 T细胞比例和表型的变化。方法采用流式细胞术来检测PV组(n=33)、PK组(n=19)及正常对照(normal control,NC)组(n=33)的外周血单个核细胞(peripheral blood mononuclear cell,PBMC)中CD3、Vγ9、Vδ2、CLA、CD27、CD45RA的表达。结果与NC组相比较,Vγ9Vδ2 T细胞占总T细胞的比例和CLA+Vγ9Vδ2 T细胞占总Vγ9Vδ2 T细胞的比例分别在PV组(t=-2.349,P=0.0224;t=-3.277,P=0.0017)和PK组(t=-2.226,P=0.0306;t=-2.901,P=0.0055)出现显著减少;Vγ9Vδ2 T细胞分化成TCM的比例在PV组和PK组分别出现明显下降(t=-4.932,P<0.0001;t=-2.572,P=0.0131);同时,TEMRA的比例在PV组和PK组均出现明显升高(t=3.849,P=0.0003;t=2.236,P=0.0353);TEM比例的下降仅在PV组具有统计学意义(t=-3.303,P=0.0018)。结论PV和PK患者的外周血Vγ9Vδ2 T细胞的比例和表型具有相似性,该细胞可能迁移至皮肤,参与皮肤的自身炎症应答。
Objective To investigate the frequencies and phenotypes of peripheral Vγ9Vδ2 T cells from patients with plaque type psoriasis vulgaris(PV)and porokeratosis(PK).Methods The expression of CD3,Vγ9,Vδ2,CLA,CD27,CD45RA were detected in PBMCs from PV group(n=33),PK group(n=19)and normal control(NC)group(n=33)using flow cytometry.Results Compared with NC group,the percentages of Vγ9Vδ2 T cells among the total T cells and CLA+Vγ9Vδ2 T cells over the Vγ9Vδ2 T cells were significantly decreased in PBMCs from patients with PV group(t=-2.349,P=0.0224;t=-3.277,P=0.0017)and PK group(t=-2.226,P=0.0306;t=-2.901,P=0.0055),respectively;the differentiation of Vγ9Vδ2 T cells into TCMwere significantly decreased in both PV and PK groups(t=-4.932,P<0.0001;t=-2.572,P=0.0131,respectively);meanwhile,the proportion of TEMRAwas significantly increased in both PV and PK groups(t=3.849,P=0.0003;t=2.236,P=0.0353,respectively);only the proportion of TEMwas significantly decreased in PV group(t=-3.303,P=0.0018).Conclusion There were similarities in the frequencies and phenotypes of peripheral Vγ9Vδ2 T cells from patients with PV and PK,those cells might be involved in the skin-homing process and autoinflammatory responses.
