接受抗逆转录病毒治疗的LTNPs外周血自然杀伤细胞功能

Function of natural killer cells in peripheral blood of LTNPs patients receiving antiretroviral therapy

目的为探讨接受抗逆转录病毒治疗的HIV-1长期不进展者(long-term non-progressors, LTNPs)与接受抗逆转录病毒治疗(antiretroviral therapy treatment, ART)的HIV进展者在自然杀伤细胞(natural killer, NK)表型与功能上的差异。方法选取LTNPs+ART及HIV进展者+ART的外周血,用流式细胞术检测CD3/CD16/CD56/CD69各表型的NK细胞比例,使用多重细胞因子试剂盒检测评估NK细胞功能的细胞因子水平。结果本研究纳入LTNPs+ART和HIV进展者+ART各38例。LTNPs+ART组CD3^(-)CD56^(+)和CD3^(-)CD16^(+) NK细胞亚组表达频率低于对照组(P=0.024、0.035);LTNPs+ART组NK细胞细胞因子(干扰素-γ、肿瘤坏死因子-α和白介素-18)水平高于HIV进展者+ART组(P均<0.01)。结论接受治疗的LTNPs表现出较低的细胞毒性,但相对比接受治疗的HIV进展者NK细胞产生的细胞因子水平较高。

Objective To illustrate the phenotype and function of natural killer(NK) cells in peripheral blood of HIV-1 long-term non-progressors(LTNPs) who received antiretroviral therapy(ART). Methods 38 LTNPs receiving ART were enrolled in the observation group, while 38 HIV progressors who also received ART were recruited in the control group.Peripheral blood samples were collected and the proportion of NK cells of each phenotype CD3/CD16/CD56/CD69 was determined by flow cytometry, and NK cell function related cytokine levels were measured using a multiplex cytokine kit. Results The CD3^(-)CD56^(+) and CD3^(-)CD16^(+) NK cell subsets were lower in the observation group than that in the control group(P=0.024, 0.035), while NK cell related cytokine levels(interferon-γ, tumour necrosis factor-α and interleukin-18) were higher, the differences were statistically significant(P<0.01). Conclusions Lower cytotoxicity is observed in LTNPs receiving ART, while the levels of cytokines produced by NK cells are higher compared to HIV progressors.
