复方鳖甲软肝片治疗肝纤维化的机制研究 被引量:4

摘要 目的筛选复方鳖甲软肝片(FFBJ)治疗肝纤维化的作用机制。方法基于网络药理学的技术和方法,本研究采用ADME/T计算方法筛选FFBJ的活性成分,依据反向药效团匹配数据库等筛选FFBJ活性成分的作用靶点,在OMIM等数据库寻找肝纤维化疾病相关靶点,映射分析筛选出FFBJ治疗肝纤维化的潜在靶点。采用Cytoscape 3.7.0软件构建活性成分-靶点网络,并对靶点基因进行基因本体(GO)富集分析和基因组百科全书(KEGG)代谢通路分析。结果共得到FFBJ 18个关键活性成分可通过20个关键靶点作用于肝纤维化,富集得到GO中92个生物过程条目和128条KEGG通路。结论FFBJ活性成分主要通过PI3K-Akt、丝氨酸蛋白激酶(MAPK)、TGF-β1等信号通路抗肝纤维化,体现了中药多成分、多靶点、多途径的作用特点,为深入开展FFBJ抗肝纤维化机制研究提供理论依据。 Objective To explore the mechanism of compound Biejia Ruangan tablets(FFBJ)in the treatment of hepatic fibrosis.Methods Based on the techniques and methods of network pharmacology,the ADME/T calculation method was used to screen the active ingredients of FFBJ,and PharmMapper database was combined to forecast and screen the targets of the main active ingredients.The related targets of hepatic fibrosis were searched from OMIM database.The mapping analysis between the targets of FFBJ and the related targets of liver fibrosis was carried out.Cytoscape 3.7.0 software was used to construct the active ingredient-disease target network of FFBJ;and the potential target of FFBJ for hepatic fibrosis was analyzed by gene ontology(GO)and genomic encyclopedia(KEGG)pathway enrichment.Results A total of 18 effective active ingredients of FFBJ were obtained,which could act on hepatic fibrosis through 20 potential targets.GO functional enrichment analysis obtained 92 GO items,and KEGG pathway enrichment analysis obtained 128 pathways.Conclusions Bioactive components of FFBJ play an anti-hepatic fibrosis role mainly through PI3K-Akt,MAPK,and TGF-β1 signal pathways,embodying the characteristics of multiple components,targets,and pathways of traditional Chinese medicine.This study provides a theoretical basis for further research on the mechanism of FFBJ against liver fibrosis.
