小麦-智利大麦染色体端体系的鉴定 被引量:1

摘要 小麦-近缘植物染色体端体系是发掘和定位小麦近缘植物优异基因的重要工具材料。为了获得小麦-智利大麦染色体端体系,本研究以寡聚核苷酸Oligo-pSc119.2-1和Oligo-pTa535-1为探针的荧光原位杂交对小麦-智利大麦1H^(ch)+1H^(ch)S附加系自交后代进行筛选,从中鉴定出小麦-智利大麦1H^(ch)S单端体系和1H^(ch)L双端体系,为将源自智利大麦1H^(ch)染色体的耐盐和抗禾谷孢囊线虫基因定位到染色体臂上提供了重要材料。为了获得可以追踪小麦背景中智利大麦1H^(ch)染色体的特异分子标记,以小麦为对照,以一套小麦-智利大麦染色体系为材料,筛选源自簇毛麦的IT引物和源自水稻的PLUG引物,结果发现,引物CINAU1186和TNAC1536可以在1H^(ch)L端体系中扩增出长度分别为350 bp和700 bp的特异多态性标记,为检测小麦中的智利大麦1H^(ch)L提供了新的检测方法。 Chromosome telosomic lines of wheat-alien species are important tool materials for exploring and locating excellent genes which derived from alien species.In order to obtain wheat-Hordeum chilense chromosome telosomic lines,the fluorescence in situ hybridization(FISH)using Oligo-pSc119.2-1 and Oligo-pTa535-1 as probes was performed on the self-pollinated progenies of wheat-H.chilense 1 Hch+1 HchS addition lines in this study.The results showed that a wheat-H.chilense 1 HchS monotelosomic line and a 1 HchL ditelosomic line were identified out,which provided important materials for locating the salt-tolerant genes or cereal cyst nematode resistant genes on chromosome arm(s)of 1 Hch.In order to establish specific molecular markers that could be used to track H.chilense 1 Hch chromosome in wheat background,PCR was performed on the genomic DNA of a set of wheat-H.chilense chromosome lines using common wheat as negative control.As a result,the IT primer CINAU1186 and the PLUG primer TNAC1536 derived from Dasypyrum villosum and Oryza sativa respectively could amplify specific polymorphic markers at the length of 350 bp and 700 bp in 1 HchL ditelosomic line,which provided new detection methods for tracing H.chilense 1 HchL in wheat background.
