摘要
肿瘤免疫逃逸是导致肿瘤恶性发展以及临床癌症免疫疗法失败的重要原因,其中肿瘤组织中的免疫细胞表达抑制性受体是某些癌症的主要逃逸机制。Siglecs受体家族由于能识别并与肿瘤细胞上的唾液酸聚糖结合,被认为与肿瘤浸润性免疫细胞的许多免疫调节作用有关。从Siglecs的角度综述目前针对Siglecs-唾液酸聚糖轴的相关研究,阐述Siglecs下调免疫细胞杀伤作用,触发免疫抑制细胞因子产生,并诱导抗肿瘤免疫反应的相关机制。此外还将讨论近年来有关Siglecs抑制剂的开发,为调节免疫反应以提高癌症免疫治疗效果提供糖生物学策略。
Tumor immune escape is an important reason for the malignant development of tumors and the failure of clinical cancer immunotherapy,in which the expression of inhibitory receptors by immune cells in tumor tissues is the main escape mechanism of some cancers.Because Siglecs receptor family can recognize and interact with sialoglycan on tumor cells,it is considered to be related with many immunomodulatory effects of tumor infiltrating immune cells.In this paper,we will review the current research on Siglecssialic acid polysaccharide axis from the perspective of Siglecs,and explain the mechanism of Siglecs downregulating the killing effect of immune cells,triggering the production of immunosuppressive cytokines and inducing anti-tumor immune response.In addition,the development of Siglecs inhibitors in recent years will also be discussed in order to provide glycobiology strategies for regulating immune response to improve the effectiveness of cancer immunotherapy.
