摘要
目的探讨XAGE-1b在肝癌细胞株中的表达与增殖侵袭的关系。方法运用荧光定量PCR检测3株肝癌细胞株XAGE-1b内源性表达情况,选取内源性表达量较高的1株细胞株进行功能实验。运用XAGE-1b干扰片段转染肝癌细胞株,荧光定量PCR检测其转染效率;运用CCK8细胞增殖实验、Boyden小室细胞侵袭实验观察肝癌细胞株体外增殖及侵袭情况。结果XAGE-1b干扰载体转染肝癌细胞株后,其XAGE-1b的mRNA表达下调(P<0.05);干扰XAGE-1b肝癌细胞体外增殖、侵袭能力下降(P<0.05,P<0.05)。结论XAGE-1b表达下调抑制肝癌细胞体外增殖及侵袭能力,初步证实XAGE-1b在肝癌增殖侵袭中发挥重要作用。
Objective To investigate the relationship between the expression of XAGE-1b and proliferation and invasion in hepatocellular carcinoma(HCC)cell lines.Methods The endogenous expression of XAGE-1b in three HCC cell lines was detected by QPCR.One cell line with the highest endogenous expression was selected for the experiment.HCC cell line was infected by XAGE-1b interference vector,and the interference efficiency was detected by QPCR.CCK8 cell proliferation assay and Boyden chamber invasion assay were used to observe the proliferation and invasion of HCC cell lines in vitro.Results The expression of XAGE-1b was down-regulation by QPCR after transfection in the HCC cell(P<0.05),the ability of proliferation and invasion were down-regulation in vitro(P<0.05,P<0.05).Conclusion Down regulation of XAGE-1b expression inhibits the proliferation and invasion of HCC cells in vitro,XAGE-1b plays an important role in the proliferation and invasion of HCC.
