Correlation between VSTM2A,MICA Genes Polymorphisms and Onset Risk of Cervical Cancer

摘要 目的探讨V-组跨膜结构域包含物2A(VSTM2A)、主要组织相容性复合体Ⅰ类相关基因A(MICA)基因多态性与宫颈癌发病风险的关联性。方法选取105例宫颈癌患者(病例组)以及105例正常健康者(对照组),对VSTM2A、MICA的单核苷酸多态性位点(SNP)rs7457728、rs2516448进行基因分型,比较病例组与对照组基因型频率,通过Logistic回归模型计算VSTM2A、MICA基因多态性与宫颈癌发病风险的关系。结果病例组VSTM2A基因的rs7457728位点中C基因频率高于对照组(P<0.05);病例组MICA基因的rs2516448位点中T基因频率高于对照组(P<0.05);病例组VSTM2A基因的rs7457728位点中CC基因型频率39.05%高于对照组9.52%,GG基因型频率14.29%低于对照组44.76%(P<0.05);病例组MICA基因的rs2516448位点中TT基因频率37.14%高于对照组22.86%,CC基因频率15.24%低于对照组26.67%(P<0.05)。Logistic回归分析显示rs7457728位点中CC型是宫颈癌发生的危险基因型,OR值为1.980(95%CI:1.325~2.959);rs2516448位点中TT型是宫颈癌发生的危险基因型,OR值为1.508(95%CI:1.029~2.210)。非鳞癌VSTM2A基因的rs7457728位点中CC基因型频率高于鳞癌(P<0.05)。结论VSTM2A基因的rs7457728位点和MICA的rs2516448位点基因多态性与宫颈癌的发病风险有关。 Objective To explore the correlation between V-set transmembrane domain containing 2A(VSTM2A),major histocompatibility complex class I chain-related gene A(MICA)genes polymorphisms and onset risk of cervical cancer.MethodsA total of 105 patients with cervical cancer(case group)and 105 normal and healthy people(control group)were enrolled.Genotyping was conducted for single nucleotide polymorphisms(SNP)of VSTM2A and MICA at rs7457728 and rs2516448 loci.The genotype frequencies were compared between the case group and the control group.The relationship between VSTM2A,MICA gene polymorphisms and onset risk of cervical cancer was calculated by Logistic regression model.Results C gene frequency of VSTM2A gene at rs7457728 locus in the case group was higher than that in the control group(P<0.05),and T gene frequency of MICA gene at rs2516448 locus was higher than that in control group(P<0.05).The CC genotype frequency of VSTM2A gene at rs7457728 locus in the case group was higher than that in the control group(39.05%vs 9.52%),while GG genotype frequency was lower than that in the control group(14.29%vs 44.76%)(P<0.05).TT gene frequency of MICA gene at rs2516448 locus in the case group was higher than that in the control group(37.14%vs 22.86%),while CC gene frequency was lower than that in the control group(15.24%vs 26.67%)(P<0.05).The results of Logistic regression analysis showed that CC type at rs7457728 locus was a dangerous genotype of cervical cancer onset[OR=1.980,95%CI:1.325~2.959)].TT type at rs2516448 locus was a dangerous genotype of cervical cancer onset[OR=1.508,95%CI:1.029~2.210].CC genotype frequency of VSTM2A gene at rs7457728 locus in patients without squamous cell carcinoma was higher than that with squamous cell carcinoma(P<0.05).Conclusion The polymorphisms of VSTM2A gene at rs7457728 locus and MICA gene at rs2516448 locus are related to onset risk of cervical cancer.
