摘要
目的探讨细胞周期数依赖性蛋白激酶4(cell cycle number dependent protein kinase4,CDK4)、CD133在骨肉瘤组织中的表达及临床意义。方法收集进行手术的骨肉瘤患者87例,在术中收集其病灶标本,选择同期进行手术的骨软骨瘤手术切除标本作为对照,采用免疫组化SP法检测2组患者病灶组织中CDK4、CD133蛋白表达情况。结果研究组病灶组织中CDK4、CD133蛋白表达阳性率(70.11%、40.66%)高于对照组(P>0.05)。骨肉瘤组织CDK4、CD133表达与外科分期、是否转移有关(P<0.05),与性别、年龄、发病部位及组织学类型无关(P>0.05)。Logrank分析结果显示,骨肉瘤组织CDK4、CD133阳性表达表达患者OS分别为26.0个月、28.0个月,低于CDK4、CD133阴性骨肉瘤患者OS的36.0个月、39.0个月(P<0.05)。结论CDK4及CD133蛋白在骨肉瘤组织中表达增加,与外科分期、肺转移及患者预后有关,其机制有待进一步研究。
Objective To explore the expression and clinical significance of cell cycle number dependent protein kinase 4(CDK4)and CD133 in tissues of patients with osteosarcoma.Methods A total of 87 patients with osteosarcoma undergoing surgery were collected.During the operation,specimens of their lesions were collected.The surgical resection specimens of osteochondroma during the same period were selected as controls.Immunohistochemical SP method was used to detect the expression of CDK4 and CD133 in the lesion tissues of the two groups Happening.Results The positive rates of CDK4 and CD133 protein expression in the lesion tissues of the study group(70.11%,40.66%)were higher than those of the control group(P>0.05).The expression of CDK4 and CD133 in osteosarcoma tissue was related to surgical staging and metastasis(P<0.05),but not related to gender,age,location of onset and histological type(P>0.05).Logrank analysis showed that the OS of patients with CDK4 and CD133 positive expression in osteosarcoma tissue was 26.0 months and 28.0 months,respectively,which was lower than the OS of 36.0 months and 39.0 months in patients with CDK4 and CD133-negative osteosarcoma(P<0.05).ConclusionThe increased expression of CDK4 and CD133 proteins in osteosarcoma tissues is related to surgical staging,lung metastasis and patient prognosis.The mechanism needs to be further studied.
