益气活血中药对良性前列腺增生模型大鼠的治疗作用及对前列腺B淋巴细胞瘤-2蛋白表达的影响 被引量:5

摘要 目的观察益气活血中药对良性前列腺增生(BPH)模型大鼠的治疗作用及对前列腺B淋巴细胞瘤-2(Bcl-2)蛋白表达的影响,并探讨其作用机制。方法将90只雄性SD大鼠随机分为假手术组(19只)和模型手术组(71只),模型手术组通过雄性激素诱导法建立BPH模型,造模结束后2组各取6只验证造模成功。再将模型手术组剩余65只大鼠随机分为模型组、非那雄胺组、中药低剂量组、中药中剂量组及中药高剂量组,每组13只,以及假手术组剩余大鼠13只,共6组。假手术组及模型组大鼠给予0.9%氯化钠注射液5 mL/d灌胃,非那雄胺组大鼠予非那雄胺片药液0.8 mg/(kg·d)灌胃,中药低、中、高剂量组每日分别予0.85、1.70、2.55 g/(kg·d)益气活血中药颗粒药液灌胃,均每日1次,连续灌胃4周。比较各组灌胃后前列腺湿质量指数变化情况,采用免疫组化法检测各组大鼠灌胃后前列腺组织中Bcl-2蛋白表达光密度值情况。结果与假手术组比较,模型组及中药低、中剂量组大鼠前列腺湿质量指数均明显增加(P<0.05);与模型组比较,非那雄胺组及中药中、高剂量组大鼠前列腺湿质量指数均明显降低(P<0.05);与非那雄胺组比较,中药低、中剂量组大鼠前列腺湿质量指数均明显增加(P<0.05);中药各剂量组组间比较,中药高剂量组大鼠灌胃后前列腺湿质量指数明显低于中药低、中剂量组(P<0.05)。与假手术组比较,模型组及中药低剂量组Bcl-2蛋白光密度值均明显升高(P<0.05);与模型组比较,非那雄胺组及中药低、中、高剂量组Bcl-2蛋白光密度值均明显降低(P<0.05);与非那雄胺组比较,中药低剂量组Bcl-2蛋白光密度值明显升高(P<0.05);中药各剂量组组间比较,中药中、高剂量组Bcl-2蛋白光密度值均明显低于中药低剂量组(P<0.05)。结论益气活血中药治疗BPH疗效确切,可明显抑制BPH模型大鼠前列腺组织增生,改善组织病理表现,降低前列腺湿质量指数,且治疗效果具有一定的量效关系,高剂量治疗效果更好,其作用机制可能与抑制前列腺组织中Bcl-2蛋白表达有关。 Objective To observe the effect of Yiqi Huoxu Granuleson on model rat with benign prostatic hyperplasia(BPH)and its effect on the expression of B lymphocyte tumor-2(Bcl-2)protein in prostate,and explore its mechanism of action.Methods Ninety healthy male SD rats were randomly divided into the sham-operated group(n=19)and the model surgery group(n=71).The model surgery group established model of BPH by androgen induction.then 6 animals from each group were taken to verify the success whether the model is successful.The remaining 65 rats in the model surgery group were randomly divided into the model group(gavaged by 5 mL/d 0.9%chloride injection),finasteride group[gavaged by 0.8 mg/(kg·d)finasteride liquid],low-dose group(gavaged by 0.85 g/(kg·d)Yiqi Huoxue Granules),medium-dose group[gavaged by 1.70 g/(kg·d)Yiqi Huoxue Granules]and high-dose group[gavaged by 2.25 g/(kg·d)Yiqi Huoxue Granules],plus sham-operated group(also gavaged by 5 mL/d 0.9%chloride injection),and 13 per group in each,a total of 6 groups.Once daily for 4 weeks in all groups,the wet mass index of prostate in each group were compared after gavaged.The optical density value of Bcl-2 protein expression in prostate tissue in each group after gavage was detected by immunohistochemistry.Results Compared with the sham-operated group,the prostate wet mass index in the model group,low-dose and medium-dose groups increased significantly after gavage(P<0.05);compared with the model group,the prostate wet mass index in the finasteride group,medium-dose and high-dose groups decreased significantly after gavage(P<0.05);compared with the finasteride group,the prostate wet mass index in the low-dose and medium-dose groups increased significantly after gavage(P<0.05);Comparison between Chinese medicine dosage group,the prostate wet mass index in the high-dose group was lower than that in the low-dose and medium-dose groups after gavage(P<0.05).Compared with the sham-operated group,the optical density value of Bcl-2 protein in the model group and low-dose group increased significantly after gavage(P<0.05);compared with the model group,the optical density value of Bcl-2 protein in the finasteride group,low-dose,medium-dose and high-dose groups decreased significantly after gavage(P<0.05);compared with the finasteride group,the optical density value of Bcl-2 protein in the low-dose group increased significantly after gavage(P<0.05);Comparison between Chinese medicine dosage group,the optical density value of Bcl-2 protein in the medium-dose and high-dose groups was lower than that in the low-dose group after gavage(P<0.05).Conclusion Yiqi Huoxue Granuleson for BPH has a definite effect,can significantly inhibit the hyperplasia of prostate tissue in BPH model rats,improve histopathological performance,decrease prostate wet mass index level,and the treatment effect has a certain dose-effect relationship.high-dose treatment is better,and its mechanism of action may be related to inhibiting the expression of Bcl-2 protein in prostate tissue.
