IGF-1通路在美洲大蠊提取物诱导胃癌细胞凋亡中的作用 被引量:3

摘要 目的探讨美洲大蠊提取物(Periplaneta americana extract)诱导MGC-803胃癌细胞凋亡的作用和作用机制。方法以胃癌MGC-803细胞为研究对象,MTT法测定细胞增殖抑制率;JC-1染色测定细胞线粒体膜电位情况;DAPI染色观察细胞核形态;流式细胞术检测细胞凋亡情况;Western blot测定凋亡相关基因bcl-2、bax和胰岛素样生长因子-1(insulin-like growth factor 1,IGF-1)信号通路相关蛋白的蛋白表达情况。结果与阴性对照组相比,50、100 mg·L-1的美洲大蠊提取物可不同程度地抑制MGC-803细胞的增殖,降低线粒体膜电位,使细胞核固缩,染色质深染,细胞凋亡增加,同时bcl-2表达降低,bax表达增加。机制研究结果显示IGF-1、磷脂酰肌醇-3激酶(phosphatidylinositol 3-kinase, PI3K)和磷酸化蛋白激酶B(protein kinase B,p-Akt)蛋白表达均有不同程度降低。结论美洲大蠊提取物对MGC-803细胞具有诱导凋亡的作用,其机制可能通过抑制IGF-1信号通路的表达实现。 Objective To investigate the effect and mechanism of Periplaneta americana extract on apoptosis of MGC-803 gastric cancer cells.Methods The effects of different concentrations of Periplaneta americana extract on human gastric cancer MGC-803 cell were observed.Proliferation inhibiting rate was measured by MTT assay.The MMP was measured by JC-1 staining.The nuclear change was analyzed by DAPI staining.Apoptosis was determined by AV/PI staining.The protein expressions of insulin-like growth factor 1(IGF-1),phosphatidylinositol 3-kinase(PI3 K),protein kinase B(Akt),p-Akt, bcl-2 and bax were detected by western blotting.Results Periplaneta americana extract(50,100 mg·L-1) dose-dependently inhibited the proliferation of human gastric cancer MGC-803 cell, decreased MMP and bcl-2 expression, induced its apoptosis and increased bax expression.Besides, the expressions of IGF-1,PI3 K,p-Akt were decreased by Periplaneta americana extract.Conclusion Periplaneta americana extract can induce apoptosis of human gastric cancer MGC-803 cell through IGF-1 signaling pathway in vitro.
