摘要
目的采用网络药理学方法筛选防风治疗肠易激综合征的作用靶点,探讨其治疗肠易激综合征的作用机制。方法通过TCMSP数据库,以口服利用度(OB)≥30%,类药性(DL)≥0.18为阈值,筛选得到防风的主要化学成分及其所作用的基因靶点。通过GeneCards数据库筛选肠易激综合征相关的基因靶点,取疾病与中药的基因靶点交集,网络合并,建立"有效成分-预测基因"对应关系,采用Cytoscape软件构建"成分-靶点-疾病"交互网络图,采用STRING数据库构建PPI网络,采用DAVID数据库进行GO功能富集分析,采用Bioconductor数据库对作用靶点进行KEGG信号通路分析,对防风治疗肠易激综合征的作用机制做出合理推测。结果通过PPI网络,共发现43个节点,319条相互作用关系;靶点基因富集的蛋白功能主要包括抗炎反应、对雌二醇反应、细胞凋亡过程的负调控等;主要信号通路有AGE-RAGE信号通路、IL-17信号通路、内分泌抵抗等。结论防风可能通过干预p38激活、核转录因子-κB转录及雌激素受体表达,缓解肠道炎症反应、高敏状态等,从而达到治疗肠易激综合征的目的。
Objective To screen the targets of Saposhnikoviae Radix in the treatment of irritable bowel syndrome by means of network pharmacology,and to investigate their mechanism of action.Methods The effective components of Saposhnikoviae Radix and the gene targets were screened out by traditional Chinese medicine systematic pharmacological(TCMSP)database,with oral bioavailability(OB)≥30%and drug-like(DL)≥0.18 selected as the thresholds.Gene targets related to irritable bowel syndrome were screened by GeneCards database.The intersection of gene targets was taken,the interaction network diagram of"ingredients-targets-diseases"was constructed using Cytoscape software.The PPI interaction analysis was conducted using STRING tool.David database was used to perform GO function enrichment analysis,and Bioconductor database was used to analyze the KEGG signal pathway of the target.Based on these results,we made a reasonable speculation on the mechanism of Saposhnikoviae Radix in the treatment of IBS.Results Through PPI network,a total of 43 nodes and 319 interaction relationships were found.The main functions involved in the targets were anti-inflammatory response,response to estradiol,negative regulation of apoptotic process,etc.,and the main enrichment pathways included AGE-RAGE signal pathway,IL-17 signal pathway,endocrine resistance,etc.Conclusion Saposhnikoviae Radix participates in p38 activation,NF-κB transcription and estrogen receptor expression in alleviate intestinal inflammation and hypersensitivity,so as to achieve the purpose of treating IBS.
