摘要
采用LC-MS/MS技术建立野生青风藤中木兰花碱和青藤碱含量的测定方法。以水-乙腈溶液为流动相梯度洗脱,以Kromasil Eternity-5 C18色谱柱(2.1 mm×50 mm,5μm)为分析柱,电喷雾离子源,正离子模式,选择离子监测(SRM)。木兰花碱和青藤碱分别在20.6~412 ng·mL^(-1)和29.5~590 ng·mL^(-1)的范围内线性良好,相关系数均大于0.99。方法专属性良好,木兰花碱和青藤碱的平均加样回收率分别为99.09%和98.95%,RSD分别为1.99%和1.65%。所建立的LC-MS/MS方法简单、灵敏,适用于野生青风藤中木兰花碱和青藤碱含量的检测。
The method for determining magnoflorine and sinomenine contents in wild Caulis Sinomenii was established by LC-MS/MS..The water-acetonitrile solution was taken as mobile phase to wash gradiently.The Kromasil Eternity-5 C18 column(2.1 mm×50 mm,5μm)was used as the analytical column.The electrospray ion source(ESI),the positive ion mode,and the selected ion monitoring(SRM)were used.Magnoflorine and sinomenine showed good linearity at ranges of 20.6~412 ng·mL^(-1) and 29.5~590 ng·mL^(-1),respectively,and the correlation coefficients were greater than 0.99.The method showed good specificity and the average recoveries of magnoflorine and sinomenine were 99.09%and 98.95%with RSD of 1.99%and 1.65%,respectively.The LC-MS/MS method is simple,sensitive and suitable for the determination of magnoflorine and sinomenine contents in wild Caulis Sinomenii.
