基于转录组的巨龙竹CesA基因家族的生物信息学分析 被引量:4

Bioinformatics Analysis of CesA Gene Family Based on the Transcriptome of Dendrocalamus sinicus

摘要 纤维素是植物细胞壁的重要组成成分,主要由多个纤维素合成酶(CesA)基因家族参与编码。本研究基于巨龙竹单分子实时测序(SMRT)转录组数据库,利用高质量转录本非冗余序列结合转录本功能注释,共筛选出45个CesA基因候选序列。巨龙竹CesA基因家族编码蛋白的氨基酸数目为351~1652个,其分子量为40.45~186.12 kD,且大部分蛋白以α-螺旋和无规则卷曲为主。通过CesA基因家族成员之间的同源性分析,以及与水稻、毛竹、绿竹和慈竹4个禾本科物种的CesA基因家族共同构建的系统进化树分析发现,巨龙竹CesA基因家族成员可分成10个类群。F01_cb9914_c103031/f5p6/3907与F01_cb9914_c4357/f4p0/4200两个基因与巨龙竹初生壁纤维素生物合成有关,其中且由共同的祖先演化而来,发生了基因功能的分化,使得巨龙竹笋的组织特异性表达存在较大差异。本研究结果对巨龙竹CesA基因家族基因的进一步研究及其功能鉴定奠定了基础。 Cellulose is the principal ingredient of cell wall in bamboo,and is mainly encoded by the cellulose synthase A(CesA)gene family.Based on the single molecule real time sequencing(SMRT)transcriptomic data of Dendrocalamus sinicus,using non-redundant high-quality transcript sequences and functional annotation,45 CesA genes were identified.that the number of amino acids in the proteins encoded by CesA gene family of D.sinicus ranged from 351 to 1652,and the molecular weight of the proteins ranged from 40.45 to 186.12 kD,and most of the proteins were mainlyα-helix and random coils.This research attempt to analyze the homology of 45 D.sinicus of CesA genes and phylogenetic tree with rice,moso bamboo,green bamboo,and Bambusa emeiensis,and these CesA gene family can be clustered into 10 class.The two genes of F01_cb9914_c103031/f5 p6/3907 and F01_cb9914_c4357/f4 p0/4200 are related to the biosynthesis of cellulose in the primary wall of the D.sinicus,both of them have a common ancestor,and gene function differentiation,which makes the tissue-specific expression of D.sinicus shoot comparatively large.The results of this research laid the foundation for the in-depth study of the CesA gene family genes and their functional identification.
