摘要
目的:从p38丝裂原活化蛋白激酶(p38 Mitogen-activated Protein Kinase,p38MAPK)炎症相关信号通路,探讨电针后血清对脂多糖(Lord Privy Seal,LPS)诱导大鼠膝关节软骨细胞退变的影响。方法:制备电针血清;酶联免疫吸附法(Enzyme-Linked Immunoassay,ELISA)检测炎性因子白细胞介素1β(Interleukin-1β,IL-1β)、肿瘤坏死因子-α(Tumor Necrosis Factor-α,TNF-α)的表达情况;反转录聚合酶链式扩增反应(Reverse Transcription-Polymerase Chain Reaction,RT-PCR)、免疫印记(Western blot)法检测电针血清干预退变软骨细胞二型胶原(collagen II)、p38MAPK、磷酸化p38丝裂原活化蛋白激酶(p-p38MAPK) mRNA和蛋白的表达。结果:电针血清可显著下调软骨细胞上清液中IL-1β、TNF-α的表达,差异具有统计学意义(P <0.05或P <0.01);RT-PCR、Western blot结果显示:电针血清可上调collagen II mRNA和蛋白含量表达,可下调p38MAPK、p-p38MAPK mRNA和蛋白含量表达,差异具有统计学意义(P <0.05或P <0.01)。结论:电针血清可通过调控collagen II、p38MAPK、p-p38MAPK mRNA和蛋白表达,在一定程度上延缓软骨细胞的退变进程。
Objective: The effects of electroacupuncture on LPS-induced the knee joint chondrocytes degeneration in rats were explored according to p38MAPK inflammation-related signal pathway. Methods: The serum after electroacupuncture was prepared, the expression of IL-1β and TNF-α were detected by ELISA. RT-PCR and Western blot methods were applied to detect the mRNA and protein expression of collagen II, p38MAPK and p-p38MAPK. Results: Electroacupuncture can significantly down-regulate the expression of IL-1β and TNF-α in the supernatant of chondrocytes(P<0.05 or P<0.01). RT-PCR and Western blot results show: Electroacupuncture can upregulate the expression of collagen II mRNA and protein expression, and down-regulate the mRNA and protein expression of p38MAPK and p-p38MAPK(P<0.05 or P<0.01). Conclusion: Electroacupuncture can delay the degeneration of chondrocytes to a certain extent by regulating the mRNA and protein expression of collagen II, p38MAPK and p-p38MAPK.
