骨巨细胞瘤中lncRNA的差异表达及其临床应用价值

摘要 目的应用基因芯片技术筛选长链非编码RNA(lncRNA)在骨巨细胞瘤患者和正常人中的差异表达,探究lncRNA对骨巨细胞瘤的早期诊断等临床应用价值。方法随机选择2016年5月至2019年5月初诊为原发骨巨细胞瘤18例患者肿瘤组织(GCTB组),18例长骨干骺端骨创伤组织(对照组),使用基因芯片技术筛选获得差异lncRNA的表达。通过GO和KEGG Pathway分析差异表达基因并使用实时荧光定量PCR(qRT-PCR)验证两组患者骨组织和血清中目标lncRNA的相对表达水平。结果选择两个具有最大差异倍数上调和下调的lncRNA,qRT-PCR结果显示,GCTB组患者骨肿瘤组织和血清中lnc-FOLR3-2:2的表达明显高于对照组,差异有统计学意义(P<0.05)。ENST00000442197.1在对照组的表达明显高于GCTB组,差异有统计学意义(P<0.05)。以两组中lnc-FOLR3-2:2和ENST00000442197.1的表达水平作为诊断指标并绘制受试者工作特征曲线(ROC),通过计算曲线下面积(AUC),lnc-FOLR3-2:2和ENST00000442197.1在诊断骨巨细胞瘤中的准确性分别为0.841和0.857。结论 Lnc-FOLR3-2:2和ENST00000442197.1在骨巨细胞瘤中表达差异,可能在该肿瘤发生及进展中发挥重要作用,为骨巨细胞瘤的早期诊断、药物治疗靶点等提供了新的方向。 Objective To screen differential expression of long-chain non-coding RNA( lncRNA) in patients with GCTB and normal people by using gene chip technology,and to explore lncRNA roles in the early diagnosis of GCTB and its clinical application value.Methods From May 2016 to May 2019,18 cases of tumor tissue diagnosed with primary GCTB and 18 cases of traumatic bone tissue from metaphysis of long bones were randomly selected that gene chip technology was used to screen separately.Differential expression of lncRNA was obtained. Differential expression genes were analyzed through GO and KEGG Pathway.Quantitative Real-time PCR( qRT-PCR) was used to verify relative expression of target lncRNA in bone tissue and serum.Results Two lncRNAs with the largest differential up-regulation and down-regulation were selected. The qRT-PCR results showed that lnc-FOLR3-2: 2 expression in tumor tissue and serum of patients with GCTB was significantly higher than that in control group.On the contrary,the ENST00000442197. 1 expression in control group was significantly higher than that in GCTB group with statistically significant difference( P<0. 05). Finally,lnc-FOLR3-2: 2 and ENST00000442197. 1 were used as diagnostic indicators that receiver operating characteristic curve( ROC) was drawn.By calculating area under ROC curve( AUC),accuracies of lnc-FOLR3-2: 2 and ENST00000442197. 1 to diagnose GCTB were 0. 841 and 0. 857,respectively. Conclusion Lnc-FOLR3-2: 2 and ENST00000442197. 1 would be significantly differential expressed in giant cell tumor of bone,which may play an important role in the occurrence and progression.It would provide a new direction for early diagnosis and drug treatment of giant cell tumor of bone.
