十八味诃子利尿丸对雄性糖尿病肾病大鼠肾组织RAGE-NF-κB信号通路的影响 被引量:6

摘要 目的探讨十八味诃子利尿丸对雄性糖尿病肾病(DN)大鼠肾组织RAGE-NF-κB信号通路的影响。方法自行设计研究方案开展相关研究。选取SD雄性大鼠10只作为对照组,20只(通过高糖高脂饮食联合链脲佐菌素尾静脉注射制备DN大鼠)作为模型组、藏药组,连续灌胃2周,用Western blot法及免疫组化法检测NF-κB P65、RAGE、TNF-α、IL-6、IL-1β,以光镜观察大鼠肾脏组织病理结构。对实验结果行相关统计学分析。结果与对照组比较,模型组肾组织NF-κB P65、RAGE、TNF-α、IL-6、IL-1β蛋白表达升高(P<0.05),病理损伤严重。与模型组比,藏药组肾组织NF-κB P65、RAGE、TNF-α、IL-6、IL-1β蛋白表达降低(P<0.05),病理损伤减轻。结论十八味诃子利尿丸能够通过抑制雄性DN大鼠肾组织RAGE-NF-κB信号通路活化,延缓DN的进展。 Objective To investigate the effects of Eighteen Flavor Myrobalan Diuretic pills of on the signaling pathway of RAGE-NF-κB in renal tissues of male rats with diabetic nephropathy.Methods Self-designed research scheme.Ten male SD rats were selected as the control group, and 20 rats(DN rats were prepared by high glucose and high fat diet combined with streptozotocin intravenous injection)as the model group and Tibetan medicine Myrobalan group.The rats were given intragastric administration for two weeks in a row.NF-κB P65,RAGE,TNF-α,IL-6 and IL-1β were detected by Western blot and immunohistochemistry.The pathological structure of renal tissues was observed by light microscopy.The experimental results were statistically analyzed.Results Compared with the control group, the protein expression of NF-κB P65,RAGE,TNF-α,IL-6 and IL-1β in renal tissues of the model group were increased(P<0.05),with serious pathological damage.Compared with the model group, the protein expression of NF-κB P65,RAGE,TNF-α,IL-6 and IL-1β in renal tissues of Tibetan medicine group were decreased(P<0.05),with alleviated pathological damage.Conclusion Eighteen Flavor Myrobalan Diuretic pills can delay the progression of DN by inhibiting the activation of signaling pathway RAGE-NF-κB in renal tissue of male rats with diabetic nephropathy.
