摘要
目的分析长链非编码RNA烟酰胺核苷酸转氢酶反义RNA1(lncRNA NNT-AS1)、微小RNA-424(miR-424)在子宫内膜癌组织中的表达及其临床意义。方法收集60例子宫内膜癌患者的子宫内膜癌组织和40例正常子宫内膜组织,检测lncRNA NNT-AS1、miR-424的相对表达水平,分析二者的相对表达水平与患者临床病理特征及预后的关系,以及二者的相关性。结果lncRNA NNT-AS1在子宫内膜癌组织中高表达,并与国际妇产科联盟(FIGO)分期、脉管浸润和淋巴结转移相关(P<0.05)。miR-424在子宫内膜癌组织中低表达,与FIGO分期、肌层浸润程度、脉管浸润和淋巴结转移相关(P<0.05)。lncRNA NNT-AS1高表达、miR-424低表达的患者5年生存率较低(P<0.05)。lncRNA NNT-AS1高表达和miR-424低表达是患者预后不良的影响因素。lncRNA NNT-AS1相对表达水平与miR-424呈负相关(r=-0.368,P<0.05)。结论lncRNA NNT-AS1和miR-424在子宫内膜癌中均异常表达,且与子宫内膜癌患者不良病理特征和预后相关。
Objective To analyze the expression and clinical significance of long non-coding RNA nicotinamide nucleotide transhydrogenase-antisense RNA1(lncRNA NNT-AS1),microRNA-424(miR-424)in endometrial carcinoma.Methods The relative expression levels of lncRNA NNT-AS1 and miR-424 were detected in the endometrial carcinoma tissues of 60 cases of endometrial carcinoma patients and 40 cases of normal endometrial tissues.The relationship between the relative expression levels and the clinicopathological features,prognosis of patients and the correlation between the two was analyzed.Results LncRNA NNT-AS1 was highly expressed in endometrial carcinoma,and it was related to International Federation of Gynecology and Obstetrics(FIGO)stage,vascular invasion and lymph node metastasis(P<0.05).MiR-424 was low expressed in endometrial carcinoma,and it was related to FIGO stage,degree of myometrial infiltration,vascular infiltration and lymph node metastasis(P<0.05).The patients with high expression of lncRNA NNT-AS1 and low expression of miR-424 had lower 5-year survival rates(P<0.05).High expression of lncRNA NNT-AS1 and low expression of miR-424 were influence factors for poor prognosis in patients(P<0.05).There was a negative correlation between the expression level of lncRNA NNT-AS1 and miR-424(r=-0.368,P<0.05).Conclusion LncRNA NNT-AS1 and miR-424 are both abnormally expressed in endometrial carcinoma,and they are associated with poor pathological features and prognosis of endometrial carcinoma.
