感染性心内膜炎与钙化性主动脉瓣疾病的相关性分析

摘要 目的采用生物信息学方法分析感染性心内膜炎(infective endocarditis,IE)患者外周血和钙化性主动脉瓣疾病(calcific aortic valve disease,CAVD)瓣膜组织的基因表达谱芯片,研究两种疾病共同差异表达基因的功能及调控网络。方法采用公共数据库(GEO)中获取的IE患者外周血和CAVD患者瓣膜组织基因表达谱芯片数据,利用R软件包分析差异表达基因,采用DAVID数据库对共同差异表达基因进行基因功能和信号通路分析。通过STRING数据库进行蛋白—蛋白相互作用网络分析,Cytoscape软件筛选最关键的主效基因(Hub基因)。结果共筛选出47个共同的差异表达基因,其功能富集生物途径主要涉及免疫应答和炎症反应,主要参与金黄色葡萄球菌感染、补体和凝血级联反应、破骨细胞的分化等信号通路。构建了共同差异表达基因的蛋白互作网络,通过对互作网络的分析筛选出了4个Hub基因SYK、FPR1、S100A9和PLEK。结论利用生物信息学方法,通过整合数据分析表明IE患者外周血中存在与CAVD相关的分子物质基础。 Objective Bioinformatics tools were used to analyze the gene expression profiles of peripheral blood from patients with infective endocarditis(IE)and aortic valve tissues from patients with Calcific Aortic Valve Disease(CAVD),and to screen the differentially co-expressed genes.Methods Peripheral blood of IE patients and valvular tissue of CAVD patients were obtained from the public database(GEO).R software package was used to analyze the differentially co-expressed genes.DAVID database was used to annotate the biological process and signaling pathway of differentially co-expressed genes.STRING database was used to analyze protein-protein interactions,and Cytoscape software was used to identify the hub genes.Results A total of 47 differentially co-expressed genes were dientified.Funtion analysis indicated that biology processes focused on the immune response and inflammation,and signaling pathway was enriched in the of Staphylococcus aureus infection,osteoclast,complement and coagulation cascades.A protein-protein interaction(PPI)network of differentially co-expressed genes was constructed.The genes such as SYK,FPR1,S100A9 and PLEK were the hub nodes of the crosstalk network.Conclusion Bioinformatic tools provides us with effective information through integrative analysis to better understand the molecular basis of interaction between IE and CAVD.
