紫癜灵颗粒对大鼠肠系膜微循环的影响

摘要 目的探讨紫癜灵颗粒改善大鼠肠系膜微循环的药效学情况。方法将50只ICR大鼠适应性喂养1周后,以10%高分子右旋糖酐6 m L/kg舌静脉给药,制备急性微循环障碍模型。将造模成功的大鼠随机分为空白组、模型组、银杏叶片组、紫癜灵高剂量组(10 g/kg)、紫癜灵中剂量组(5 g/kg)、紫癜灵低剂量组(2.5 g/kg),每组10只。受试药物用蒸馏水配制成混悬液,灌胃给药15 m L/kg,空白组和模型组给予同等体积的蒸馏水。观察紫癜灵颗粒对大鼠肠系膜急性微循环障碍微动脉流速、微静脉流速、微静脉流态、微动脉口径、微静脉口径、毛细血管开放数的影响。结果与空白组比较,模型组在5、15、30、45、60min的微动、静脉流速减慢,模型组在15、30、45、60 min的微静脉流态加权积分升高,模型组在5、15、30、45、60 min的微动、静脉口径缩小,模型组在5、15、30、45、60 min的毛细血管开放数减少,差异均有统计学意义(P<0.05);与模型组比较,银杏叶片组、紫癜灵颗粒高、中剂量组在15、30、45、60 min的微动脉流速加快,银杏叶片组、紫癜灵颗粒高剂量组在5、15、30、45、60 min的微静脉流速加快,银杏叶片组、紫癜灵颗粒高、中剂量组在15、30、45 min的微静脉流态加权积分降低,银杏叶片组、紫癜灵颗粒高、中剂量组在15、30、45 min的微动脉口径增加,紫癜灵颗粒高剂量组在15、30、45、60 min的微动脉口径增加,银杏叶片组、紫癜灵颗粒高、中剂量组在15、30、45、60 min的微静脉口径增加,银杏叶片组、紫癜灵颗粒高、中、低剂量组在30、45 min的毛细血管开放数增加,差异均有统计学意义(P<0.05)。结论紫癜灵颗粒具有显著改善微循环的作用,紫癜灵颗粒能够明显加快肠系膜微循环细动、静脉血流速度,扩张细动、静脉血管管径,增加毛细血管网开放数目。 Objective To investigate the pharmacodynamics of Zidianling granule in improving mesenteric microcirculation in rats.Methods 50 ICR rats were fed adaptively for one week,and then treated with 10%high molecular dextran 6 ml/kg via lingual vein.The rats were randomly divided into blank group,model group,Ginkgo biloba group,high-dose group(10 g/kg),medium dose group(5 g/kg)and low-dose group(2.5 g/kg),with 10 rats in each group.The test drug was prepared into suspension with distilled water and given 15 ml/kg by gavage.The blank group and model group were given the same volume of distilled water.The effect of Zidianling granule on the flow velocity of micro artery,micro vein,flow pattern of micro vein,caliber of micro artery,caliber of micro vein and opening number of capillary in rats with acute mesenteric microcirculation disorder were observed.Results Compared with the blank group,the micro motion and venous flow velocity of the model group decreased at 5,15,30,45 and 60 min,the weighted integral of micro venous flow pattern of the model group increased at 15,30,45 and 60 min,the micro motion and venous caliber of the model group decreased at 5,15,30,45 and 60 min,and the capillary opening number of the model group decreased at 5,15,30,45 and 60 min,The differences were statistically significant(P<0.05).Compared with the model group,the flow velocity of the micro artery in the high-dose and middle dose groups of Ginkgo biloba and Zidianling granules increased at 15,30,45 and 60 min,the flow velocity of the micro vein in the high-dose group of Ginkgo biloba and Zidianling granules increased at 5,15,30,45 and 60 min,the flow weighted integral of the micro vein in the high-dose and middle dose groups of Ginkgo biloba and Zidianling granules decreased at 15,30 and 45 min,and the flow weighted integral of the micro vein in the high-dose and middle dose groups of Ginkgo biloba and Zidianling granules decreased The diameters of arterioles in high-dose group and middle dose group increased at 15,30 and 45 min,those in high-dose group increased at 15,30,45 and 60 min,those in Ginkgo biloba group and high-dose group and middle dose group increased at 15,30,45 and 60 min,those in high-dose group and low-dose group increased at 30,30,45 and 60 min The number of capillaries opened at 45 min increased,and the differences were statistically significant(P<0.05).Conclusion Zidianling granule can significantly improve microcirculation.Zidianling granule can significantly accelerate the blood flow velocity of mesenteric microcirculation arterioles and veins,expand the diameter of arterioles and veins,and increase the number of capillary network opening.
