一株桑椹核盘菌的分离鉴定及植物源抑菌剂的筛选 被引量:4

Isolation and Identification of a Sclerotinia sclerotiorum Strain from Mulberry Fruit and Screening of Its Plant-Derived Bacteriostatic Agents

摘要 采集陕西省周至县桑园土壤中生长的疑似核盘菌子实体,通过组织分离法从子实体分离到一株真菌命名为Z02。菌落形态和分子生物学鉴定结果显示Z02为核盘菌(Sclerotinia sclerotiorum)。对该菌进行生物学特性研究发现,在试验条件下该菌生长最适pH为3,最适温度为15℃,果糖和葡萄糖为利用效果较好的碳源,酵母粉为利用效果较好的氮源。使用菌丝生长速率法筛选对其具有抑制作用的植物提取物,结果显示八角茴香的60%醇提取液具有抑菌作用,当供试浓度为0.1 g/mL时,抑菌率为97.21%。 Suspected fruiting body of Sclerotinia sclerotiorum was collected from soil of mulberry field in Zhouzhi County,Shaanxi Province,and a fungus named Z02 was isolated from the fruiting body by tissue isolation method.The morphological and molecular biological identification showed that Z02 was a Sclerotinia sclerotiorum strain.According to the test on its biological characteristics,the optimal growth pH was 3 and optimal growth temperature was 15℃under laboratory condition.The suitable carbon source was fructose and glucose,and suitable nitrogen source was yeast powder.Then,plant extracts which had bacteriostatic effect on Z02 were screened out by mycelial growth rate method.The results showed that 60%alcohol extract of star anise had antibacterial effect.When the concentration of plant extract was 0.1 g/mL,the antibacterial rate was 97.21%.
