微小RNA-376b-3p和微小RNA-654-5p在大肠癌中的表达及临床意义

Expression and clinical significance of microRNA-376b-3p and microRNA-654-5p in colorectal cancer

目的本研究旨在探讨微小RNA(miR)-376b-3p和miR-654-5p在大肠癌中的表达水平及其临床意义。方法荧光定量PCR检测34组来自2018年6月至2019年1月淮安市肿瘤医院大肠癌及癌旁组织中miR-376b-3p和miR-654-5p的表达。利用肿瘤基因组图谱(TCGA)数据库分析miR-376b-3p和miR-654-5p的表达与大肠癌患者临床病理因素以及生存预后的关系。应用SPSS 17.0软件对所有结果进行统计分析。结果相对于癌旁组织,大肠癌组织中miR-376b-3p表达下调,miR-654-5p表达上调,差异有统计学意义(P<0.001)。miR-376b-3p表达的降低和miR-654-5p表达的升高与肿瘤的转移和临床分期密切相关。此外,单因素分析结果显示miR-376b-3p的低表达或miR-654-5p的高表达的大肠癌患者总体生存率较低,差异有统计学意义(P<0.05)。多因素分析发现miR-376b-3p低表达和miR-654-5p高表达是大肠癌患者预后差的独立预测因子。结论miR-376b-3p的低表达和miR-654-5p的高表达可能与大肠癌的发生、发展明显相关。

Objective To investigate the expression levels and clinical significance of miR-376b-3p and miR-654-5p in colorectal cancer.Methods Fluorescence quantitative PCR was used to detect the expression of miR-376b-3p and miR-654-5p in 34 cases of colorectal cancer and adjacent tissues from June 2018 to January 2019 in Huai’an Tumor Hospital.The relationship between the expression of miR-376b-3p and miR-654-5p and the clinicopathological factors and survival prognosis of patients with colorectal cancer was analyzed using the cancer genome atlas(TCGA)database.Statistical analysis of all results was done by the SPSS 17.0 software.Results The expression of miR-376b-3p was down-regulated and that of miR-654-5p was up-regulated in colorectal cancer tissues(P<0.01).The decrease of miR-376b-3p expression and the increase of miR-654-5p expression were closely related to tumor metastasis and clinical stage.The univariate analysis showed that the overall survival rate of colorectal cancer patients with low expression of miR-376b-3p or high expression of miR-654-5p was low(P<0.05).The multivariate analysis revealed that the low expression of miR-376b-3p and high expression of miR-654-5p were independent predictors of poor prognosis in patients with colorectal cancer.Conclusion The low expression of MiR-376b-3p and the high expression of miR-654-5p may be related to the occurrence and development of colorectal cancer.MiR-376b-3p and miR-654-5p can be used as independent prognostic markers for colorectal cancer.
