基于转录组测序构建食管腺癌预后相关的内源竞争RNA网络

摘要 目的挖掘食管腺癌中内源竞争RNA(ceRNAs)网络,探索预后影响机制。方法提取癌症和肿瘤基因图谱计划(TCGA)数据库中正常食管组织和食管腺癌组织转录组数据,挖掘差异表达且与预后相关的长链非编码RNA(lncRNA)、短链非编码RNA(miRNA)及mRNA。采用权重基因共表达网络分析预后相关基因的关联性,构建ceRNA网络。采用基因集富集分析(GSEA)探索影响预后的可能机制。结果ACO87388.1、AC131009.3、miR-29c-3p、CDK1、TPX2是在正常组织和食管腺癌组织中预后相关的差异表达基因,并可构成ACO87388.1/miR-29c-3p/CDK1、ACO87388.1/miR-29c-3p/TPX2、AC131009.3/miR-29c-3p/CDK1、AC131009.3/miR-29c-3p/TPX2四种ceRNA调控网络。结论ACO87388.1、AC131009.3、miR-29c-3p、CDK1和TPX2是影响食管癌患者预后的重要分子。 Objective To explore the network of competing endogenous RNAs(ceRNAs)in prognosis of esophageal adenocarcinoma.Methods The transcriptome data of normal esophageal tissues and esophageal adenocarcinoma tissues in The Cancer Genome Atlas(TCGA)database were extracted to mine the differentially expressed long non-coding RNA(lncRNA),microRNA(miRNA)and mRNA related to prognosis of esophageal adenocarcinoma.Weighted gene co-expression network analysis was used to analyze the prognostic related genes and to construct a ceRNA network for prognosis of esophageal adenocarcinoma.Gene set enrichment analysis(GSEA)was conducted to explore the possible mechanisms that affect prognosis.Results ACO87388.1,AC131009.3,miR-29c-3p,CDK1,TPX2 were differentially expressed genes between normal tissues and esophageal adenocarcinoma tissues,which could constitute ACO87388.1/miR-29c-3p/CDK1,ACO87388.1/miR-29c-3p/TPX2,AC131009.3/mregulatory iR-29c-3p/CDK1 and AC131009.3/miR-29c-3p/TPX2 ceRNA networks for prognosis of esophageal adenocarcinoma.Conclusion ACO87388.1,AC131009.3,miR-29c-3p,CDK1 and TPX2 are important molecules,which may be associated with the prognosis of patients with esophageal cancer.
