摘要
目的:研究青蒿琥酯(ART)对2型糖尿病(T2DM)大鼠下颌下腺中IL-1β表达的影响。方法:将24只健康雄性SD大鼠随机分为正常对照组、模型组和ART组。模型组和ART组给予高糖高脂饲料喂养,8周后腹腔注射35 mg/kg链脲佐菌素(STZ)复制T2DM模型。用药干预4周后,观察各组大鼠体重、空腹血糖、唾液流量的变化,分别采用实时荧光定量PCR(qPCR)法和免疫组化法检测大鼠下颌下腺中IL-1β基因和蛋白的表达。结果:与模型组相比,ART组大鼠血糖显著降低,体重显著增加,唾液流量明显增加(P<0.05)。模型组IL-1βmRNA及蛋白表达较正常对照组显著上调,ART组大鼠IL-1βmRNA及蛋白表达较模型组显著下调(P<0.05)。结论:ART能降低T2DM大鼠血糖,增加其唾液流量,减轻下颌下腺损伤,其机制可能与抑制下颌下腺中IL-1β的表达有关。
Objective:To investigate the effect of artesunate(ART)on the expression of IL-1βin the submandibular gland of type 2 diabetic(T2DM)rats.Methods:Twenty-four healthy male SD rats were randomly divided into control group,model group and ART group.Model group and ART group rats were fed with high-sugar and highfat diet,and were intraperitoneally injected with 35mg/kg streptozotocin(STZ)8 weeks later to replicate the T2DM model.After 4 weeks of drug intervention,the changes of body weight,fasting blood glucose and salivary flow in each group were observed.The expression of IL-1βin submandibular gland was detected by real-time qPCR and immunohistochemical method.Results:Compared with model group,the blood glucose was significantly decreased while the body weight and salivary flow were notably increased in ART group(P<0.05).Compared with the control group,the mRNA and protein expression of IL-1βin the model group wasevidently up-regulated,and that was significantly down-regulated in the ART group compared with the model group(P<0.05).Conclusion:ART can reduce blood glucose,increase salivary flow and alleviate submandibular gland injury in T2DM rats.The mechanism may be related to the inhibition of IL-1βexpression in the submandibular glands.
