电针百会和四神聪对缺血再灌注损伤模型鼠学习记忆能力及细胞凋亡的影响 被引量:8

Effects of Electroacupuncture at Baihui and Sishencong Acupoints on Learning and Memory Ability and Apoptosis in MCAO Model Rats

摘要 目的:观察电针百会穴、四神聪穴对中动脉闭塞(MCAO)模型鼠学习记忆能力的改善作用,并探讨其相关机制。方法:将80只SD大鼠随机分为假手术组(n=20)、造模组(n=60),造模组大鼠接受改良线栓法制备缺血再灌注损伤模型,将其中成模的40只随机分为模型组、电针组及抑制剂(LY294002)组,每组10只。假手术组、模型组大鼠仅接受模拟抓取,1次/d,电针组及抑制剂组大鼠接受电针百会穴、四神聪穴,1次/d,30 min/次,连续干预14 d,用Morris水迷宫测大鼠学习记忆行为学能力改变,用2,3,5-氯化三苯四唑(TTC)染色法观察大鼠脑梗死体积变化,Tunel检测神经元细胞凋亡情况,用Western Bloting检测PI3K、AKT、p-AKT水平变化,用酶联免疫吸附试验法(ELISA)检测大鼠外周血清Bcl-2、Bax的水平变化。结果:电针四神聪可明显改善MCAO大鼠学习记忆能力,减少脑梗死体积及神经元的凋亡率,降低Bax水平,上调PI3K、p-AKT、Bcl-2水平,上述作用可被PI3K-AKT信号通路抑制剂LY294002削弱。结论:电针百会穴、四神聪穴可改善缺血再灌注损伤所致的学习记忆障碍,其作用机制可能是通过PI3K-AKT信号通路实现。 Objective:To observe the effect of electroacupuncture at Baihui Acupoint and Sishencong Acupoint on learning and memory ability of MCAO model rats and explore its related mechanism.Methods:A total of 80 Sprague Dawley(SD)rats were randomly divided into a sham operation group(n=20)and a model group(n=60).Modified suture method was used to establish the Middle cerebral artery occlusion(MCAO)model of ischemia-reperfusion injury in the model group.A total of 40 rats in the model group whose model was established successfully were randomly divided into a model group,an electroacupuncture group,and an inhibitor(LY294002)group,with 10 rats in each group.Rats in sham operation group and model group only received simulated capture once a day.Rats in electroacupuncture group and inhibitor group received electroacupuncture at Baihui acupoint and Sishencong acupoint once a day for 14 days,with 30 minutes for each time.The behavioral changes of learning and memory were measured by Morris water maze;the changes of cerebral infarction volume were observed by TTC staining,and the changes of neuronal cell apoptosis were detected by Tunel.Western Blot was used to detect the changes of PI3K,AKT and p-AKT.The levels of Bcl-2 and Bax in peripheral serum of rats were detected by ELISA.Results:Electroacupuncture at Sishencong Acupoint could significantly improve the learning and memory ability of MCAO rats,reduce the volume of cerebral infarction and the apoptosis rate of neurons,reduce the level of Bax,and up-regulate the levels of PI3K,p-AKT and Bcl-2,which could be weakened by LY294002,an inhibitor of PI3K-AKT signal pathway.Conclusion:Electroacupuncture at Baihui Acupoint and Sishencong Acupoint can improve the learning and memory impairment caused by ischemia-reperfusion injury,and its mechanism may be realized through PI3K-AKT pathway.
