结合WGCNA构建与猪肌肉脂肪酸相关的调控网络

摘要 试图通过加权基因共表达网络分析(WGCNA)构建与猪脂肪酸性状相关的关键调控网络,旨在为提高猪肉质品质提供信息。基于猪种质资源发掘与创新利用-山西省科技创新重点团队前期猪肌肉组织的转录组测序数据进行分析,采用WGCNA划分模块,Cytoscape软件插件Clue GO对重要模块进行富集分析并筛选重要模块,利用Cytoscape软件可视化共表达网络。结果表明,分析聚类得到15个模块,对各个模块进行富集分析,发现MEgreen模块和MEyellow模块与脂肪酸的分解代谢通路、PPAR信号通路等显著相关,这些通路均在脂肪酸组成中有重要作用。分别从每个模块筛选出10条核心基因,MEgreen模块核心基因为NDUFB7、CPT2、NDUFB3、ATP5O、NDUFAB1、CPOX、ATP5A1、MRM1、NDUFV1和ETFA,模块中还有7条lncRNAs;MEyellow模块核心基因为PSTPIP2、PGM1、UBE2D4、PGAM2、WWP1、DENND5A、ACTR1B、MACROD1、CAND2和PYGM,模块中还有9条lncRNAs。研究采用WGCNA获得了影响猪脂肪酸组成的关键因子,结果可为解析猪肉质性状形成的分子机制提供理论参考。 In this study,weighted gene co-expression network analysis(WGCNA)was used to construct a regulatory network related to fatty acid composition in pigs,aiming to provide information for improving pork quality.Based on the transcriptome sequencing data of pig muscle tissue in the earlier stage of Shanxi Swine Germplasm Resources Exploitation&Innovation Utilization Key Team,WGCNA was used to divide the modules,Cytoscape software plug-in ClueGO was used to enrich and screen the important modules,and Cytoscape software was used to visualize the co-expression network.The result showed that 15 modules were obtained.The enrichment analysis of each module showed that MEgreen module and MEyellow module were significantly correlated with the catabolism pathway and PPAR signaling pathway of fatty acids,which all played an important role in fatty acid composition.10 hub genes were screened from each module.The hub genes of MEgreen module were NDUFB7,CPT2,NDUFB3,ATP5O,NDUFAB1,CPOX,ATP5A1,MRM1,NDUFV1 and ETFA,and there were 7 lncRNAs in the module.The hub genes of MEyellow module were PSTPIP2,PGM1,UBE2D4,PGAM2,WWP1,DENND5A,MACROD1,CAND2,ACTR1B and PYGM,and there were 9 lncRNAs in the module.WGCNA was used to obtain the hub molecules affecting the fatty acid composition of pigs.The results will provide theoretical reference for the molecular mechanism of the formation of pork quality traits.
