摘要
为了研究抗冻剂组合和平衡时间与鸡精液冻后活力的关系,试验选用8%甘油、8%甘油+6%二甲基亚砜(DMSO)、8%甘油+6%DMSO+4%乙二醇(EG)3个抗冻剂组合与基础液Ⅰ液(渗透压约为330 mOsm,pH值为7.0)组成冷冻稀释液(Ⅱ液),加鸡精液平衡0,30,60,90,120,150,180,210 min,测定不同平衡时间下鸡精液冻后活力和精子复苏率。结果表明:抗冻剂8%甘油和8%甘油+6%DMSO均能提高鸡精液冻后活力,且解冻后精子活力随着平衡时间的延长呈现先升高后降低的趋势,而抗冻剂组合8%甘油+6%DMSO+4%EG使鸡精液冻后活率呈现下降趋势;用8%甘油、8%甘油+6%DMSO、8%甘油+6%DMSO+4%EG分别作为抗冻剂稀释精液,在精液稀释后平衡时间分别为60,30,0 min时,其解冻后精子活力依次为0.4167,0.4333,0.4167,均明显高于其他平衡时间;精子复苏率分别为52.09%、54.16%和52.09%;存活时间分别为169,145,89 min。说明添加抗冻剂组合8%甘油+6%DMSO的稀释精液在平衡30 min时,精液解冻后精子活力最高,复苏率最高,存活时间较长。
In order to study the relationship between the combination equilibration time of cryoprotectant and the vitality of chicken semen after freezing,three cryoprotectant combinations(8%Glycerol,8%Glycerol+6%DMSO,8%Glycerol+6%DMSO+4%EG)were selected in this experiment,and separately combined with the extenderⅠ(Osmotic pressure 330 mOsm,pH value 7.0)into a freezing dilute(Ⅱ).The chiken semen was added for balancing 0,30,60,90,120,150,180 and 210 min,the sperm viability and recovery rate after freezing were measured.The results showed that:Both 8%Glycerol and 8%Glycerol+6%DMSO all can improve chicken semen-s frozen-thawed motility,but the semen-s frozen-thawed motility was promoted firstly and then decreased with the extension of the equilibrium time.As the solute used 8%Glycerol+6%DMSO+4%EG as cryoprotectant,the semen-s frozen-thawed motility took on the tendency of decline gradually with the extension of the equilibrium time.When the semen was diluted with 8%Glycerol,8%Glycerol+6%DMSO,and 8%Glycerol+6%DMSO+4%EG as cryoprotectant,the equilibration time of the solutes were 60 min,30 min and 0 min,respectively;the sperm motility after thawing was 0.4167,0.4333,and 0.4167,respectively,which was significantly higher than other equilibration time;the recovery rate of semen was 52.09%,54.16%and 52.09%,respectively,the semen-s survival time was 169 min,145 min and 89 min,respectively.The results indicated that the sperm motility,recovery rate and survival time were the highest after thawing when the diluted semen with 8%glycerol+6%DMSO was balanced for 30 min.
