芪枝渴痹通方对高糖诱导大鼠雪旺细胞RSC96活性氧含量及凋亡的影响

Effect of Qizhi Kebitong Recipe on the reactive oxygen species content and apoptosis of rat Schwann cells RSC96 induced by high glucose

目的探讨芪枝渴痹通方对高糖诱导的大鼠雪旺细胞(RSC96)活性氧(ROS)含量及凋亡率的影响。方法使用高糖诱导RSC96细胞,建立DPN细胞模型,MTT法检测细胞存活率,流式细胞术检测ROS含量及凋亡率。结果不同浓度的葡萄糖对RSC96细胞的存活率呈剂量依赖性降低(P<0.05,P<0.01,P<0.001);不同浓度的甘露醇对RSC96细胞的存活率无影响;50、100、150μg·mL^(-1)浓度的芪枝渴痹通方对RSC96细胞具有增殖作用(P<0.05,P<0.001);芪枝渴痹通方可提高高糖诱导RSC96细胞存活率(P<0.01,P<0.001);芪枝渴痹通方可降低高糖诱导RSC96细胞ROS含量及细胞凋亡率(P<0.01,P<0.001)。结论芪枝渴痹通方可显著降低高糖诱导RSC96细胞ROS含量并抑制细胞凋亡率。

Objective To investigate the effect of Qizhi Kebitong Recipe on the content of reactive oxygen species(ROS)and apoptosis rate of rat Schwann cells(RSC96)induced by high glucose.Methods RSC96 cells were induced with high glucose to establish a DPN cell model.MTT assay was used to detect the cell survival rate,and flow cytometry was used to detect the ROS content and apoptosis rate.Results Different concentrations of glucose reduced the survival rate of RSC96 cells in a dose-dependent manner(P<0.05,P<0.01,P<0.001),while different concentrations of mannitol had no effect on the survival rate of RSC96 cells.Qizhi Kebitong Recipe with concentrations of 50,100 and 150μg·mL^(-1) had a proliferation effect on RSC96 cells(P<0.05,P<0.001).It increased the survival rate of RSC96 cells induced by high glucose(P<0.01,P<0.001),and reduced the ROS content and apoptosis rate of RSC96 cells induced by high glucose(P<0.01,P<0.001).Conclusion Qizhi Kebitong Recipe can significantly reduce the ROS content of RSC96 cells induced by high glucose and inhibit the rate of cell apoptosis.
