摘要
目的改进《中国药典》2020年版葶苈子中槲皮素-3-O-β-D-葡萄糖基-7-O-β-D-龙胆双糖苷含量测定的方法。方法用30%甲醇水回流提取,色谱柱为Agilent TC-C 18柱(250 mm×4.6 mm,5μm),流动相为乙腈-0.1%醋酸水溶液(11∶89),柱温30℃,流速为1.0 mL·min^(-1),检测波长为254 nm。结果槲皮素-3-O-β-D-葡萄糖基-7-O-β-D-龙胆双糖苷在0.120~2.367μg(r=0.99999),平均回收率(n=9)为99.13%,RSD为0.2%。10批样品测定结果分别为0.116%,0.089%,0.095%,0.106%,0.101%,0.093%,0.138%,0.087%,0.105%,0.099%。改进方法测得的含量约是《中国药典》方法的4倍。结论该方法灵敏、简便、准确,可用于葶苈子的质量控制。
OBJECTIVE To improve the determination method of Quercetin-3-O-β-D-glucose-7-O-β-D-gentiobioside in Descurainiae Semen by Chinese Pharmacopoeia 2020.METHODS Useing 30%methanol for the refluxing extraction.The separation was performed on an Agilent TC-C 18 column(4.6 mm×250 mm,5μm).The mobile phase was consisited of acetonitrile-0.1%p hosphoric acid(11∶89)at 30℃column temperature.The flow rate was 1.0 mL·min^(-1).The detection wavelength was 254 nm.RESULTS Querc etin-3-O-β-D-glucosyl-7-O-β-D-gentiobioside showed a good linear relationship at the ranges of 0.120-2.367μg(r=1.00000).The average recovery was 99.13%,the RSD was 0.2%.10 batchs of sample were determined respectively,the concertration were 0.116%,0.089%,0.095%,0.106%,0.101%,0.093%,0.138%,0.087%,0.105%and 0.099%,respectively.The determination results by this optimized method were quadrupled of Chinese Pharmacopoeia method.CONCLUSION The method was sensitive,simple,accurate,and suitable for the quality control of Descurainiae Semen.
