EPO和CEPO对心肌梗死大鼠梗死面积和血管新生的作用的比较

Effects of EPO and CEPO on Infarct Size and Angiogenesis in Rats with Myocardial Infarction

摘要 目的:探讨氨甲酰促红细胞生成素(CEPO)和促红细胞生成素(EPO)对心肌梗死大鼠梗死面积和新生血管的作用。方法:购置SD大鼠40只为研究对象,随机取SD大鼠10只设为空白对照组,剩余30只SD大鼠采用左冠状动脉结扎诱导大鼠发生急性心肌梗死动物模型,建模后随机分为模型对照组、EPO组和CEPO组。空白对照组和模型对照组在相同时期给予相同剂量的生理盐水耳缘静脉注射,CEPO组耳缘静脉注射CEPO(每周2次,单次注射剂量为50ug/kg),EPO组耳缘静脉注射EPO(注射剂量为0.3mg/kg),比较各组梗死面积、血管新生数量、血流动力学参数。结果:CEPO组干预后1个月LVSP、+dp/dtmax、-dp/dtmax水平均高于模型对照组和EPO组(P<0.05);LVDP水平低于模型对照组和EPO组(P<0.05);CEPO组干预后1个月梗死面积均小于模型对照组和EPO组(P<0.05);EPO组干预后1个月梗死面积小于模型对照组(P<0.05);免疫组化结果表明:EPO组干预后1个月血管新生数量多于模型对照组但少于CEPO组(P<0.05)。结论:EPO和CEPO均能减少心肌梗死大鼠面积,但是后者效果更佳,能改善病灶部位血流动力学水平,可能与血管新生有关。 Objective:To investigate the effects of carbamoyl erythropoietin(CEPO)and erythropoietin(EPO)on infarct size and neovascularization in rats with myocardial infarction.Methods:40 Sprague Dawley rats were purchased and 10 SD rats were randomly selected as blank control group.The remaining 30 SD rats were randomly divided into model control group,EPO group and CEPO group after acute myocardial infarction modeling induced by ligation of left coronary artery.The blank control group and model control group were given the same dose of normal saline by ear vein at the same time,CEPO was injected into ear edge vein in CEPO group(twice a week,single injection dose was 50ug/kg),EPO was injected into ear edge vein in EPO group(injection dose was 0.3mg/kg).The infarct area,number of angiogenesis and hemodynamic parameters of each group were compared.Results:The levels of LVSP,+dp/dtmax and-dp/dtmax in CEPO group were higher than those in model control group and EPO group(P<0.05).LVDP level was lower than that in model control group and EPO group(P<0.05).The infarct area of CEPO group was smaller than that of model control group and EPO group at 1 month after intervention(P<0.05).The infarct area of EPO group was smaller than that of model control group at one month after intervention(P<0.05),the number of angiogenesis in EPO group was more than that in model control group,but less than that in CEPO group(P<0.05).Conclusion:Both EPO and CEPO can reduce the area of myocardial infarction in rats,but the latter has better effect and can improve the hemodynamic level of the lesion,which may be related to angiogenesis.
