响应面法优化水龙多酚提取工艺及其抗氧化活性研究 被引量:3

摘要 目的采用响应面法优化水龙多酚的提取工艺并研究其抗氧化活性。方法以水龙多酚提取量为检测指标,在单因素实验基础上运用Box-Behnken响应面法设计四因素三水平提取实验;对优选的提取方法得到的水龙多酚进行抗氧化活性测定。结果最佳工艺条件为:超声时间40 min、超声功率550 W、乙醇浓度40%、料液比1:26(g/mL),在此条件下水龙多酚的提取率为(28.55±0.21)mg/g,其DPPH自由基清除IC50为39.1μg/mL, ABTS自由基清除IC50为108.1μg/mL。结论提取工艺优化后,水龙多酚提取率增加,得到的水龙多酚具有较好的抗氧化活性。 Objective To optimize the ultrasonic assisted extraction of polyphenol from Ludwigia adscendens(L.) Hara by response surface methods, and study the antioxidant activity of the extracted polyphenol. Methods With polyphenol content from Ludwigia adscendens(L.) Hara as index, the Box-Behnken response surface method was used to design the four-factor and three-level extraction experiments based on the single-factor experiment. Antioxidant activity of the extracted polyphenol was tested by the DPPH free radical assay and ABTS? assay. Results The best extraction process of polyphenol was as follows: the ultrasonic time was 40 min, the ultrasonic power was 550 W, the ethanol concentration was 40% and the solid-liquid ratio was 1:26(g/mL). Under these conditions, the yield of polyphenol was(28.55±0.21) mg/g. The IC50 of DPPH free radical scavenging activity was 39.1 μg/mL and the IC50 of ABTS·scavenging activity was 108.1 μg/mL. Conclusion After the optimization of extraction process, the extraction rate of polyphenol increases, and the obtained polyphenol has good antioxidant activity.
