摘要
目的研究宫颈癌患者癌组织与非癌组织中骨髓来源抑制性细胞(MDSCs)和调节性T细胞(Tregs)的频率及功能。方法收集宫颈癌患者的宫颈组织标本,分为癌组织和非癌组织两组,使用胰酶消化法处理成单细胞悬液,使用流式细胞仪分析两组之间MDSCs和Treg细胞的频率和功能的差异。结果宫颈癌患者癌组织中单核细胞型(Mo)-MDSCs和Treg细胞比例明显高于非癌组织(P<0.01),且Mo-MDSCs的一氧化氮(NO)水平在癌组织中显著升高(P<0.05)。此外,在540d的随访中,宫颈癌组织中Mo-MDSCs水平升高与无进展生存期呈显著正相关(P=0.0372)。结论Mo-MDSCs和Treg细胞在宫颈癌免疫应答中发挥着免疫抑制作用,且宫颈癌组织内高频率的Mo-MDSCs对患者的预后产生不利影响。
Objective To study the frequency and function of myeloid-derived suppressor cells(MDSCs)and regulatory T cells(Tregs)in cancerous and non-cancerous tissues of patients with cervical cancer.Methods Cervical tissue samples from patients with cervical cancer were collected and divided into cancerous and non-cancerous tissues.Trypsin digestion was applied for preparation of single cell suspension.Flow cytometry was conducted to analyze the difference in frequency and function of MDSCs and Tregs cells between the two groups.Results The frequencies of Mo-MDSCs and Tregs cells in cervical cancer tissues were significantly higher than that in non-cancer tissues,and the NO level of Mo-MDSCs in cancer tissues was significantly higher than that of non-cancer tissues(P<0.05).In addition,during the 540 follow-up days,the accumulation of Mo-MDSCs in cervical cancer tissue was positively correlated with progression-free survival of patients(P=0.0372).Conclusion Mo-MDSC can play a promoting role in the immune response of cervical cancer and accelerates the development of tumor.In addition,Mo-MDSC has an adverse effect on the prognosis of cervical cancer.
