摘要 免疫检查点分子B7-H3在多种类型的恶性肿瘤中异常高表达,普遍与肿瘤的不良愈后相关,参与了肿瘤的免疫逃逸。最近研究表明B7-H3在促进肿瘤发生和转移中起到关键作用,并证实B7-H3是肿瘤细胞增殖、迁移、侵袭、上皮-间充质转变、肿瘤干性、耐药和Warburg效应的关键启动子。B7-H3在肿瘤免疫应答中的调控机制尚不清楚,但针对B7-H3的免疫检查点抑制剂或联合使用其他抑制剂能够明显增强免疫细胞特异性识别肿瘤细胞的能力。本文分析了B7-H3结构特征、表达特点及其调控肿瘤细胞蛋白表达的机制,并对其在肿瘤细胞增殖、侵袭、代谢以及B7-H3抗体在肿瘤免疫治疗中的作用的研究进展进行了综述。B7-H3作为新的肿瘤治疗靶点将成为具有前景的免疫治疗研究方向。 B7-H3 is highly expressed in various types of tumors and is generally associated with poor recovery of the tumor,and is also involved in immune evasion of the tumor.Recent studies have shown that B7-H3 plays a key role in promoting tumor genesis and metastasis,and confirmed that B7-H3 is a key promoter of tumor cell proliferation,migration,invasion,epithelial-mesenchymal transition,tumor dryness,drug resistance and Warburg effect.Although the mechanism for the regulation of B7-H3 in tumor immune response is unclear,studies have shown that immunocheckpoint inhibitors targeting B7-H3 or in combination with other inhibitors can significantly enhance the specific recognition of tumor cells by immune cells.In this paper,the structural characteristics and expression characteristics of B7-H3 and the mechanism of B7-H3 regulating protein expression in tumor cells were summarized.The research progress of B7-H3 in tumor cell proliferation,invasion,metabolism and the role of B7-H3 antibody in tumor immunotherapy was reviewed.As a new target of tumor therapy,B7-H3 is considered to be a promising research direction of immunotherapy.
