染矽尘大鼠脂质代谢组学的研究

摘要 目的对染矽尘大鼠血浆及肺泡灌洗液(BAL)中脂质进行脂质组学分析,探讨染矽尘大鼠有意义的脂质组学变化。方法健康雄性SD大鼠40只随机分为生理盐水对照组(NC组)和染矽尘模型组(Si组)。Si组通过气管非暴露插管灌注法注入二氧化硅(SiO2)。第42天后,麻醉大鼠留取血浆样本及肺泡灌洗液,进行质谱分析。脂质代谢学数据进行了两组间逐项t检验、主成分分析、Logistic回归分析。结果 Si组血浆及肺泡灌洗液中肉豆蔻酰溶血磷脂酸(14:0LPA)、磷脂酰溶血磷脂酸(22:6LPA)、亚油酰溶血磷脂酸(18:2LPA)的含量较NC组增高,差异有统计学意义(P<0.01);Si组血浆及肺泡灌洗液中棕榈酰溶血磷脂酰乙醇胺(16:0LPE)、硬脂酰溶血磷脂酰乙醇胺(18:0 LPE)、油酰溶血磷脂酰乙醇胺(18:1 LPE)、亚油酰溶血磷脂酰乙醇胺(18:2 LPE)、月桂酰溶血磷脂酰胆碱(12:0 LPC)、棕榈酰溶血磷脂酰胆碱(16:0 LPC)、硬脂酰溶血磷脂酰胆碱(18:0 LPC)及油酰溶血磷脂酰胆碱(18:1 LPC)的含量较NC组减低,差异有统计学意义(P<0.01)。主成分分析结果显示,占主成分70%以上的脂质为血14:0LPA、血18:0LPC、血16:0LPC。Logistic回归得到回归模型为-90.556XLPE181,预测的准确度为100%。结论血LPE18:1、14:0LPA、18:0LPC及16:0LPC为有意义的染矽尘大鼠脂质组学变化。 Objective For looking for the valuable lipid omics data of silicosis, lipid metabolomics analysis was performed in plasma and alveolar lavage fluid(BAL)on silica treated rats. Method Healthy male SD rats were randomly divided into normal saline control group(NC group)and silica treated group(Si group). The Si group was make model by injecting silica(SiO2)through the trachea. Plasma samples and BAL were collected after 42 days. The data of lipid analysis was determinated by mass spectrometry. The statistical analysis of t test between the two groups, principal component analysis, logistic regression analysis was performed. Results The contents of plasma and BAL of Myristyl lysophosphatidic acid(14:0 LPA),phosphatidyl lysophosphatidic acid(22:6 LPA),linolysin lysophosphatidic acid(18:2 LPA)in the Si group were significantly higher than those of the NC group(P<0.01).The contents of plasma and BAL of palmitoyl hemolysis phosphatidyl ethanolamine(16:0 LPE),stearyl hemolysis phosphatidyl ethanolamine(18:0 LPE),oleoyl hemolysis phosphatidyl ethanolamine 18:1(LPE),oleoyl hemolysis phosphatidyl ethanolamine(LPE),certain lauroyl hemolysis phosphatidyl choline(12:0 LPC),palmitoyl hemolysis phosphatidyl choline(16:0 LPC),stearyl hemolysis phosphatidyl choline(18:0 LPC),oleoyl hemolysis phosphatidyl choline(18:1)was significantly lower than those of the NC group(P<0.01). The results of principal component analysis showed that the lipid in blood, which was accounted for more than 70% of the principal component, was 14:0 LPA,18:0 LPC,16:0 LPC. Logistic regression obtained a regression model of-90.556 XLPE181,and the prediction accuracy of logistic regression equation was 100%. Conclusion LPE18:1,14:0 LPA,18:0 LPC and 16:0 LPC were significant lipid omics changes in silica treated rats.
