PIEZO1基因SNP与抗阻训练效果敏感性关联研究 被引量:3

Analysis of the Correlation Between PIEZO1 Gene SNP and Resistance Training’s Effects

摘要 目的:分析PIEZO1基因SNP对抗阻训练干预效果的影响,筛选抗阻训练效果敏感的基因标记。方法:招募非规律运动健身受试者190名,对其进行3个月70%1RM强度的抗阻训练(2次/周),分别在抗阻训练干预前、干预后测量深蹲1RM、纵跳指标。取受试者静脉血,提取DNA并进行基因分型,使用PLINK软件将SNP位点与抗阻训练干预效果进行关联分析,显著性水平定为0.05;根据每个位点不同基因型对表型的贡献,赋予这些位点不同的分数,得到总基因型易感分数(TGPS),将TGPS与表型之间进行相关性分析;GTEx(V8)数据库验证位点变异与基因表达差异之间的关联性。结果:1)PIEZO1 rs4433802与深蹲1RM之间显著相关(P<0.05),其中GG型对抗阻训练更敏感。2)在GTEx数据库中发现rs4433802变异与骨骼-肌肉组织中PIEZO1基因表达水平高度相关,GG基因型PIEZO1表达量显著高于AG/AA型。3)PIEZO1rs2242166、rs11861910、rs57816951、rs35957746与纵跳最大高度之间显著相关(P<0.05),这4个位点总基因型易感分数(TGPS)与表型进行相关性分析,存在弱相关关系(r2=0.153,P<0.05)。结论:1)12周70%1RM抗阻训练可以显著改善肌肉力量和功能。2)PIEZO1 rs4433802、rs2242166、rs11861910、rs57816951和rs35957746位点会影响抗阻训练改善肌肉力量和功能的效果,rs4433802 GG基因型对抗阻训练后1RM的增长更敏感;rs2242166 GG/GT、rs11861910 CC、rs57816951 CC和rs35957746 GG基因型对抗阻训练后纵跳最大高度的增加更敏感,这些位点可以作为预测抗阻训练效果的遗传分子标记。 This study aims to analyze the impact of PIEZO1 gene SNP on the outcome of resistance training and screen relevant genetic markers.Research methods:190 non-regular exercise subjects are recruited to receive resistance training of 70%1 RM for 3 months(2 times/week);before and after the resistance training intervention,they are required to take the 1 RM and vertical jump testing.Their venous blood samples are collected;DNA is extracted and genotyped.The PLINK software is used to analyze the correlation between SNPs and the effects of resis-tance training.Depending on the contribution of different genotypes to the phenotype,different scores are assignedto these loci to obtain the total genotypic predisposition score(TGPS);the GTEx(V8)database is used to furtherverify the correlation between these loci and phenotype.Results:1)A significant correlation(P<0.05)is observedbetween rs4433802 and 1 RM,and the GG type is more sensitive to resistance training.2)In the GTEx database,the rs4433802 variant is found to be highly correlated with PIEZO1 gene expression levels in skeletal muscle tis-sues,with the GG genotype having significantly higher PIEZO1 expression than the AG/AA genotypes;3)Signifi-cant correlations are found between rs2242166,rs11861910,rs57816951,rs35957746 and maximum height ofCMJ(P<0.05),and there is a weak correlation between TGPS and phenotype(r2=0.153,P<0.05).Conclusions:1)Resistance training with 70%1 RM for 12 weeks significantly improves muscle strength and function;2)rs4433802,rs2242166,rs11861910,rs57816951 and rs35957746 influence the effects of resistance training;rs4433802 GG genotype is more sensitive to the increase in 1 RM;rs2242166 GG/GT,rs11861910 CC,rs57816951 CC and rs35957746 GG genotypes are more sensitive to the improvement of maximal height of CMJ,and these SNPs can be used as genetic molecular markers to predict the effects of resistance training.
