高效液相色谱-串联质谱研究呋喃妥因代谢物在鲫鱼体内的残留消除规律

Determination of Residues and Elimination of Nitrofurantoper Metabolites in Cavassius auratus by High Performance Liquid Chromatography-Tandem Mass Spectrometry

摘要 采用HPLC-MS/MS研究呋喃妥因代谢物1-氨基乙内酰脲(AHD)在鲫鱼体内的残留消除规律。在22±1℃的水温下,模拟自然养殖条件,用质量浓度为100 ng·mL^(-1)的呋喃妥因药浴鲫鱼12 h,停止药浴后,对鲫鱼体内残留的AHD进行测定,并对鲫鱼内脏样品的提取进行除脂优化。研究结果显示,内脏经正己烷除脂后,加标回收率明显升高,为89.5%~110.2%;AHD在鲫鱼体内的残留量随着消除时间的增加呈现降低的趋势,鱼体肌肉中平均消除速率0.012μg·kg^(-1)·h,内脏中的平均消除速率为1.47μg·kg^(-1)·h,内脏的消除速率明显高于肌肉;鲫鱼肌肉和内脏中AHD的残留量分别在144 h和220 h后低于检测限0.25μg·kg^(-1)。 The residual elimination of nitrofurantoin metabolite 1-aminohydantoin(AHD)in Cavassius auratus was studied by HPLC-MS/MS.The natural aquaculture conditions were simulated,and the squid in the nitrofurantoin bath at a concentration of 100 ng·mL^(-1) was used for 12 h at a water temperature of 22±1℃After stopping the medicated bath,the residual AHD in the carp was measured,and degreasing optimization of sample extraction from squid.The results showed that after the viscera was degreased by n-hexane,the recovery rate of the spiked label was significantly increased,ranging from 89.5%to 110.2%;The residual amount of AHD in the carp showed a decreasing trend with the elimination time,the average elimination rate in the fish muscle was 0.012μg·kg^(-1)·h,and the average elimination rate in the viscera was 1.47μg·kg^(-1)·h,significantly higher than muscle.Residues of AHD in muscle and viscera of carp were lower than the detection limit of 0.25μg·kg^(-1)after 144 h and 220 h,respectively.
