^(60)Co-γ射线辐照灭菌升降茶适用性研究

摘要 目的:优选^(60)Co-γ射线辐照灭菌升降茶的最佳剂量,考察灭菌效果。方法:采用不同剂量0 kGy(空白组)、4 kGy(低剂量组)、6 kGy(中剂量组)、8 kGy(高剂量组)^(60)Co-γ射线辐照灭菌,比较灭菌前和灭菌后性状、显微特征、薄层色谱、水分变化;并运用SPSS 20.0软件分析灭菌前和灭菌后需氧菌总数、霉菌酵母菌总数、主成分(姜黄素、芦荟大黄素、大黄酸、大黄素、大黄酚、大黄素甲醚)含量变化。结果:最佳辐照剂量为中剂量(6 kGy)。与空白组比较,中剂量组性状、显微特征、薄层色谱无变化;水分无明显影响(RSD%<2);需氧菌总数、霉菌酵母菌总数均降低,有统计学差异(P<0.05),且符合《中国药典》2015年版标准规定,控制菌均为阴性;主成分含量无统计学差异(P>0.05)。结论:^(60)Co-γ射线辐照可用于升降茶的灭菌,最佳灭菌剂量为6 kGy。 Objective:The best dose of ^(60)Co-γirradiation sterilization of Shengjiang tea was optimized,and the sterilization effect was investigated.Methods:Different doses 0 kGy(blank group),4 kGy(low dose group),6 kGy(medium dose group),8 kGy(high dose group)of ^(60)Co-γirradiation sterilization were used.The changes of properties,microscopic characteristics,thin layer chromatography and water content before and after sterilization were compared.The total number of aerobic bacteria,the total number of mold yeast and the content change of principal component(curcumin,aloe emodin,rhein,emodin,chrysophanol,physcion)before and after sterilization were analyzed by SPSS 20.0 software.Results:The best irradiation dose was the medium dose(6 kGy).Compared with the blank group,the properties,microscopic characteristics,thin layer chromatography of medium dose group had no change;water content had no significant effect(RSD%<2);the total number of aerobic bacteria and the total number of mold yeast were all lower,which had significant differences(P<0.05),and met the requirements of Chinese Pharmacopoeia(2015 Edition).The control bacteria was all negative.There was no significant difference in the content of principal components(P>0.05).Conclusion:^(60)Co-γirradiation can be used for sterilization of Shengjiang tea,and the best sterilization dose is 6 kGy.
