Nesfatin-1对高糖诱导足细胞损伤的保护作用

摘要 目的探讨Nesfatin-1对高糖诱导足细胞损伤的保护作用。方法将足细胞MPC-5分为对照组、高糖组、治疗组(高糖+Nesfatin-1),倒置显微镜观察各组MPC-5细胞形态并用CCK-8检测其存活率,Western blot检测MPC-5细胞中Nephrin的表达变化。结果显微镜下观察可见,对照组足细胞呈多边形,由胞体向外伸出足突,而25 mmol·L^(-1)高糖条件下可见足细胞足突出现回缩,细胞大部分呈鹅卵石状,细胞胞体较对照组变大。而给予Nesfatin-1后,细胞胞体较高糖组变小,形态有向多边形转化的趋势,接近对照组。高糖组与对照组存活率比较,高糖诱导足细胞存活率降低(P<0.05),而Nesfatin-1处理后与高糖组相比较改善甚至促进足细胞存活(P<0.05)。Western blot结果显示与对照组相比,足细胞标记物Nephrin表达在高糖组降低,在Nesfatin-1治疗组有所升高(P均<0.05)。结论Nesfatin-1可减轻高糖诱导的足细胞损伤。 Objective To investigate the protective effect of Nesfatin-1 on podocyte injury induced by high glucose.Methods The podocyte MPC-5 cells were divided into control group,high glucose group and treatment group(high glucose+Nesfatin-1).The morphology of MPC-5 cells was observed by inverted microscope and the survival rate was observed by CCK-8.The expression of Nephrin in MPC-5 cells was detected by Western blot.Results Under the microscope,the podocytes in the control group were polygonal,protruding from the cell body,while under the condition of high 25 mmol·L^(-1) glucose,the podocyte retracted,most of the cells were pebble-shaped,and the cell body became larger than that of the control group.After the addition of Nesfatin-1,the cell body of the high glucose group became smaller,and the morphology tended to be polygonal,which was close to that of the control group.Compared with the control group,the survival rate of podocytes induced by high glucose decreased in the high glucose group,while the podocyte survival rate was improved or even promoted after Nesfatin-1 treatment compared with the high glucose group(P<0.05).The Western blot results showed that compared with the control group,the expression of Nephrin,podocyte marker,was significantly decreased in the high glucose group,while increased in the Nesfatin-1 treatment group(P<0.05).Conclusion Nesfatin-1 can alleviate podocyte injury induced by high glucose.
