摘要
目的探讨槲皮素益生元及抑菌作用,为开发利用槲皮素提供理论依据。方法采用活菌计数法和比浊法测定并分析不同浓度槲皮素对肠道10种常见菌株生长增殖的影响,用气相色谱仪测定培养上清中短链脂肪酸的含量,用MTT法分析槲皮素对Caco-2细胞活性的影响。结果在模拟胃肠道环境中不同浓度槲皮素可促进嗜酸乳酸杆菌CGMCC1.1878和鼠李糖乳酸杆菌CGMCC1.3724的生长(P<0.05),可显著提高这两种菌株丙酸和丁酸生成量(P<0.05),并且可显著(P<0.05)抑制粪肠球菌ATCC29212、大肠杆菌CMCC21531、鼠伤寒沙门氏菌CMCC50015、金黄色葡萄球菌CMCC26003四种致病菌的生长增殖。两种乳酸杆菌菌体槲皮素干预组与其无处理组比较Caco-2细胞的活性显著增强(P<0.05)。结论槲皮素可选择性的影响肠道菌株的生长增殖,有很好的益生元及抑菌作用。
Objective To investigate the prebiotic and antibacterial effects of quercetin,and provide data for the application of quercetin.Methods Colony counting and turbidimetry method were used to determine the effect of quercetin on the growth of ten common intestinal strains.Gas chromatography was used to detect the content of short-chain fatty acids in the culture supernatant,and the MTT method was employed to explore the effect of the quercetin treated bacteria and culture supernatant on the viability of Caco-2 cells.Results Quercetin significantly(P<0.05)promoted the growth of Lactobacillus acidophilus CGMCC1.1878 and Lactobacillus rhamnosus CGMCC1.3724,and increased their propionic acid and butyric acid production while significantly(P<0.05)inhibiting the growth of Enterococcus faecalis ATCC29212,Escherchia coli CMCC21531,Salmonella typhimurium CMCC50015 and Staphylococcus aureus CMCC26003.The viability of Caco-2 cells was 1.5-2 times higher than that of the control group after co-culture with two Lactobacillus strains treated with quercetin.Conclusion Quercetin can selectively affect the growth of some bacterial strains,and have prebiotic effects.
