摘要
向已建立的牙鲆 (Paralichthysolivaceus)鳃细胞系[1] (FG 930 7)中接种淋巴囊肿病 (Lymphocystisdiseasevirus ,LCDV)、探讨其是否可支持LCDV的增殖。光学显微镜观察发现 :接种 3天后 ,呈纤维状的鳃细胞隆起、立体感加强 ;之后细胞逐渐变圆 ,一周左右绝大部分鳃细胞变圆、呈现了细胞病变 (cytopathiceffect ,CPE)现象。将CPE细胞制作超薄切片 ,电镜观察发现细胞内有大量具囊膜、直径 133~ 186nm的LCDV。CPE细胞的核变形、线粒体增加且空洞化 ,出现了与淋巴囊肿细胞相同的特征。本实验证明 :FG 930 7是可支持LCDV增殖的鱼类细胞系。
Lymphocystis disease Virus(LDCV) is inoculated into cell line FG-9307, trying to explore whether it can support proliferation of LDCV. Observation of photo-microscope showed that, three days after inoculation, fibriform gill cell bulged and showed increased stereoscopy; afterward, cell became ellipse gradually; about one week after inoculation, most gill cell become ellipse, show apparent CPE. Electron-microscope observation of super-thin slice of CPE cell showed that there are lots of LCDV with envelopes whose diameters range from 133 to 186nm, the structure of cell also has changed, and the characterization of LCDV cell has appeared. All those results indicated that FG-9307 supports the proliferation of LCDV.
