杜仲全基因组GRAS基因家族的鉴定和系统发育分析 被引量:3

摘要 为研究杜仲中GRAS基因对杜仲生长发育及抗逆性的作用机制,本研究首先利用软件TBtools、Pfam和NCBI-CDD对杜仲GRAS基因进行了筛选和验证,随后借助ExPASy在线软件分析了杜仲GRAS蛋白的理化性质并通过软件MEGA 7.0构建了杜仲GRAS基因的系统进化树,最后使用在线软件MEME 5.2获得了杜仲GRAS基因的基因保守基序并通过Muscle程序和Jalview软件分析了杜仲GRAS蛋白的VHIID结构域特征。结果显示,杜仲中GRAS基因家族共含有分属于13个亚族的47个成员,其蛋白长度在388~803之间,蛋白分子量在42987~91063.2之间,等电点在4.49~9.67之间,并且都具有典型的GRAS结构域。通过对杜仲中的GRAS基因家族成员进行全面系统地分析,发现EuGRAS基因具有基因结构上的保守性和差异性,这可能与杜仲的环境适应性有关,这为进一步开展有关杜仲环境适应性机制并将其应用于濒危植物的保护等方面的研究奠定了基础。 In order to study the mechanism of the GRAS gene in Eucommia ulmoides on the growth and development and stress resistance of Eucommia ulmoides.This study used the software TBtools,Pfam and NCBI-CDD to screen and verify the GRAS genes of Eucommia ulmoides first,and then used ExPASy online software analyzed the physicochemical properties of Eucommia GRAS protein and constructed the phylogenetic tree of Eucommia GRAS gene through the software MEGA7.0.Finally,the online software MEME5.2 was used to obtain the gene conserved motif of Eucommia GRAS gene and passed the Muscle program and Jalview.The software analyzes the characteristics of the VHIID domain of Eucommia GRAS protein.The results showed that the GRAS gene family in Eucommia ulmoides contains 47 members belonging to 13 subfamilies,with protein lengths ranging from 388 to 803,protein molecular weights ranging from 42987 to 91063.2,and isoelectric point at 4.49~9.67,and all have typical GRAS domains.Through a comprehensive and systematic analysis of the members of the GRAS gene family in Eucommia ulmoides,it is found that EuGRAS genes are conservative and different in gene structure.This may be related to the environmental adaptability of Eucommia ulmoides.It would lay the foundation for the research for further development of environmental adaptation on the mechanism of sex and its application in the protection of endangered plants.
