2019新型冠状病毒潜在膜融合蛋白在雄性小鼠生殖系统的表达 被引量:1

摘要 目的研究生殖系统2019新型冠状病毒膜融合蛋白mRNA的表达情况。方法选取2020年5月从中山大学实验动物中心购买的6只12周龄C57小鼠作为研究对象。取正常雄性小鼠肾脏、睾丸、附睾头部、附睾体部、附睾尾部、输精管、精囊、前列腺及阴茎等组织,提取RNA,RT-PCR检测ACE2、CD147、TMPRSS2、ADAM17、IFITM3、LY6E、GPI的mRNA表达量。结果 ACE2在睾丸、附睾头部、精囊及阴茎有表达;CD147在生殖系统各部位均有较高表达;TMPRSS2在各部位的表达量均较少;ADAM17,IFITM3,LY6E,GPI在睾丸、附睾、输精管、精囊及前列腺均有不同程度的表达;ADAM17在睾丸、附睾头、精囊及阴茎表达量较高;IFITM3在睾丸、附睾头、附睾尾、精囊、前列腺及阴茎表达量较高;LY6E在附睾、输精管及前列腺表达量较高;GPI在各部表达均高,以睾丸、附睾尾及阴茎表达量为著。结论 2019新型冠状病毒膜融合蛋白mRNA在雄性小鼠生殖系统呈现不同水平表达,提示男性生殖系统亦存在受该病毒侵害的潜在危险。 Objective To investigate the mRNA expression of SARS-CoV-2 membrane fusion protein in reproductive system. Methods Six 12-week-old C57 mice purchased from the Experimental Animal Center of Sun Yat-sen University in May 2020 were selected as the research objects. RNA was taken from the tissues of kidney, testis, caput epididymis, corput epididymis, cauda epididymis, vas deferens, seminal vesicle, prostate and penis of normal male mice, and mRNA expression levels of ACE2, CD147, TMPRSS2, ADAM17, IFITM3, LY6 E and GPI were detected by RT-PCR. Results ACE2 was expressed in tissues of testis, caput epididymis. CD147 was expressed at a high level in each of these tissue. The expression of TMPRSS2 was low in all parts. ADAM17, IFITM3, LY6 E and GPI were expressed at a different level in testis, epididymis, vas deferens, seminal vesicle and prostate. ADAM17 was highly expressed in testis, caput epididymis, seminal vesicle, prostate and penis. IFITM3 was highly expressed in testis, caput epididymis, cauda epididymis, seminal vesicle, prostate and penis. LY6 E was highly expressed in epididymis, vas deferens and prostate. GPI was highly expressed in each of these tissues, expecially in testis, cauda epididymis, and penis. Conclusions The mRNAs of membrane fusion proteins of SARS-CoV-2 were expressed at different levels in the reproductive system of male mice, suggesting that the male reproductive system also has the potential risk of being damaged by this virus.
