miR-17-5p靶向PI3K/AKT信号通路调控皮肤鳞状细胞癌细胞增殖、凋亡的机制 被引量:10

摘要 目的探讨miR-17-5p靶向磷脂酰肌醇3-激酶/蛋白激酶B(PI3K/AKT)信号通路调控皮肤鳞状细胞癌细胞增殖、凋亡的机制。方法实时荧光定量(qRT)-聚合酶链式反应(PCR)检测HaCaT和A431细胞中miR-17-5p的表达。构建抑制miR-17-5p表达A431细胞株,噻唑蓝(MTT)法检测细胞增殖活力,流式细胞术检测细胞凋亡,Western印迹检测PIK3R1、p-AKT、p-PI3K、细胞周期素(Cyclin)D1、Bax和Bcl-2蛋白的表达。采用双荧光素酶报告基因法和Western印迹验证miR-17-5p和PIK3R1的靶向关系。结果qRT-PCR检测结果显示A431细胞中miR-17-5p的表达显著高于HaCaT(P<0.05)。MTT法、流式细胞术和Western印迹结果证实在抑制miR-17-5p表达的A431细胞中,p-AKT、p-PI3K、CyclinD1和Bcl-2的表达明显下调,PIK3R1和Bax的表达显著上调,细胞的增殖活力减弱,凋亡率增加(均P<0.05)。PI3K/AKT信号通路激活剂胰岛素样生长因子(IGF)-1可以逆转抑制miR-17-5p表达对A431细胞的增殖抑制和凋亡促进作用。结论miR-17-5p通过靶向下调PIK3R1激活PI3K/AKT信号通路促进皮肤鳞状细胞癌细胞的增殖,抑制细胞凋亡。
