Sh-RNAi对NG108-15细胞膜受体基因TNF-R1的干扰

摘要 目的NG108-15细胞膜肿瘤坏死因子(TNF)受体(TNF-R)1在受到基因干扰后,阿黑皮素原(POMC)、神经肽(NP)Y和刺鼠相关肽(AGRP)神经内分泌基因受到干扰的情况。方法采用sh-RNAi基因干扰技术干扰TNF-R1的基因表达,并利用RT-PCR技术检测TNF-R1、POMC、NPY和AGRP基因的表达情况。结果针对TNF-R1受体的膜外半胱氨酸富集区1(CRD1)所设计的sh-1干扰序列,对TNF-R1受体的基因表达具有明显的下调作用,并进而对POMC、NPY和AGRP等神经内分泌基因产生程度不同的下调影响。结论可通过sh-RNAi干扰技术来治疗TNF-α诱发的下丘脑神经内分泌性疾病。
