原发性皮肤淀粉样变中的单核/巨噬细胞数量及功能研究 被引量:1

摘要 目的观察原发性皮肤淀粉样变(PCA)患者单核/巨噬细胞在体内外的数量和功能变化。方法共纳入PCA患者20例,健康对照15例。使用流式细胞术检测外周血中CD14+单核细胞比例,免疫组织化学观察真皮中巨噬细胞数量,使用透射电镜观察巨噬细胞状态,通过Dextran(右旋糖酐)摄取实验评估体外培养的单核细胞来源的巨噬细胞(MDMs)吞噬能力,流式细胞术检测巨噬细胞极化表型。使用两样本独立t检验、单因素方差分析进行统计学分析。结果PCA患者外周血CD14^(+)单核细胞数量正常,皮损局部巨噬细胞增多,电镜下可见真皮内巨噬细胞尝试吞噬淀粉样团块。在离体环境下,PCA患者MDMs吞噬功能和极化表型与正常人没有差异。结论PCA患者外周血以及真皮中单核/巨噬细胞的数量并未减少。在离体环境下,患者的巨噬细胞未表现出吞噬功能和极化表型的异常。 Objective To observe the number and function of monocytes/macrophages of PCA patients.Methods Twenty patients with PCA and 15 healthy volunteers were enrolled in this study.Flow cytometry was used to analyze the proportion of CD14+monocytes in peripheral blood.Immunohistochemistry was performed to examine the quantity of macrophages in dermis.Transmission electron microscopy was used to observe the status of macrophages in PCA lesions and normal skin.The phagocytic function of macrophages was examined by the dextran uptake experiment.The independent sample t-test and one-way ANOVA were used for statistical analysis.Results The number of CD14^(+) monocytes in the peripheral blood of PCA patients was normal.The infiltration of macrophages in the PCA lesions increased.The intradermal macrophages were observed trying to phagocytize amyloid masses when viewed by electron microscopy.The phagocytic function and polarization phenotype of MDMs in PCA patients cultured in vitro were not different from those of normal individuals.Conclusion The number of monocytes/macrophages in the peripheral blood and dermis of PCA patients dose not decrease.The patient's macrophages dose not show abnormalities in phagocytic function and polarization phenotype.
