香菇液体菌种培养基及摇瓶培养条件优化 被引量:7

摘要 建立液体菌种优质高效生产技术体系是香菇工厂化生产的重要保障。以香菇‘森源16’为试验菌株,选择菌丝生物量、菌丝球密度和直径为主要评价指标,采用单因素和正交试验L9(34)优化香菇摇瓶液体发酵工艺。结果:棉秆粉为最佳碳源,麸皮为最佳氮源;优化培养基配方为葡萄糖3 g/100mL,棉秆粉1.5 g/100mL,木屑粉1.5 g/100mL,麸皮0.6 g/100mL,KH_(2)PO_(4)0.1 g/100mL,MgSO_(4)·7H_(2)O 0.05 g/100mL;最优摇瓶发酵条件为装液量100 mL/250mL,摇床转速170 r/min,初始pH 5.5,羧甲基纤维素钠0.20 g/100mL,漆酶0.05 g/100mL。 The establishment of a high-quality and high-efficiency production technology system for liquid spawn is an important guarantee for the factory production of Lentinula edodes.In this study,Senyuan No.16 was used as the test strain,and the mycelial biomass,mycelial ball density and diameter were used as the main evaluation indicators.Single factor test and orthogonal test L9(34)were used to optimize the liquid culture process of L.edodes in shaking flasks.Results showed that cotton stalk powder is the best carbon source and wheat bran is the best nitrogen source.The optimized medium formula is glucose 3 g/100 mL,cotton stalk powder 1.5 g/100 mL,wood chip powder 1.5 g/100 mL,wheat bran 0.6 g/100 mL,KH_(2)PO_(4)0.1 g/100 mL and MgSO_(4)·7 H_(2)O 0.05 g/100 mL.The optimal culture conditions for shake flasks are 100 mL/250 mL of liquid volume,170 r/min of shaker speed,initial pH of 5.5,sodium carboxymethyl cellulose 0.25 g/100 mL,laccase 0.05 g/100 mL.
