XRCC2在结直肠癌中的表达及其临床意义 被引量:1

Expression and Clinical Significance of XRCC2 in Colorectal Carcinoma

摘要 目的通过对300例结直肠癌病例的研究,分析结直肠癌患者中XRCC2的表达以及临床意义。方法应用免疫组化和qRT-PCR的方法,检测癌组织、癌旁组织中的XRCC2表达水平,探讨其与病理及预后的关系。结果癌组织XRCC2水平较癌旁组织更高(P<0.05);免疫组化提示癌组织XRCC2阳性者占比69.67%;qRT-PCR检测提示XRCC2在癌组织中的表达水平(1.7±0.3)显著高于癌旁组织(0.5±0.1)(P=0.0001)。XRCC2表达与Duke’s分期、TNM分期、T分期、癌灶大小、M分期、肝脏转移、淋巴结转移相关(P<0.05)。XRCC2水平较低患者的无复发生存期、总生存期更长(P<0.05)。回顾性Cox分析提示结直肠癌预后因素与Duke’s分期、XRCC2、淋巴结转移、年龄有关(P<0.05)。结论在结直肠癌患者中,XRCC2表达水平偏高,建议临床将其作为预测、判断结直肠癌患者病情进展、预后的主要标志。 Objective In this study,we investigated 300 cases of colorectal cancer to analyse the expression and clinical signifi-cance of XRCC2(Cross complementary gene 2 for X-ray repair).Methods XRCC2 expression level in cancer tissues and adjacent tissues were detected by immunohistochemistry and qRT-PCR to further explore its relationship with pathology and prognosis.Re-sults The XRCC2 level in cancer tissues was higher than that in adjacent tissues(P<0.05).Immunohistochemistry indicated that XRCC2 was positive in 69.67%of cancer tissues,qRT-PCR detection indicated that XRCC2 expression level in cancer tissues(1.7±0.3)(P=0.0001)was significantly higher than in adjacent tissues(0.5±0.1).XRCC2 expression was correlated with Duke's stage,TNM stage,T stage,tumor size,M stage,liver metastasis,lymph nodes metastasis(P<0.05).Patients with low XRCC2 level have longer overall survival and recurrence-free survival(P<0.05).Retrospective Cox analysis suggested that prognostic factors of colorectal cancer were associated with Duke's stage,XRCC2,lymph node metastasis,and age(P<0.05).Conclusion XRCC2 expres-sion level is high in patients with colorectal cancer.It is suggested that XRCC2 should be used as the main marker to predict the progress and prognosis of patients with colorectal cancer.
