葛根素通过抗氧化应激对缺血性脑损伤小鼠的神经保护作用 被引量:7

Neuroprotective effect of puerarin on ischemic brain injury in mice by antioxidant stress

摘要 目的观察葛根素通过抗氧化应激对抗缺血性脑损伤小鼠的神经保护作用。方法根据体重将小鼠随机分为4组:假手术组、模型组和低、高2个剂量实验组,每组各15只。造模之前,低、高2个剂量实验组小鼠每天分别腹腔注射葛根素(50,100 mg·kg^(-1)),连续4 d。于第5天夹闭小鼠双侧颈总动脉缺血30 min后,进行再灌注,建立缺血性脑损伤模型。低、高2个剂量实验组小鼠继续连续给药物8 d。假手术组与模型组小鼠给予生理盐水。用筑巢实验评价小鼠的日常活动自理能力,用Y迷宫实验评价小鼠的空间工作记忆能力,用试剂盒检测小鼠的前额叶皮质脑内丙二醛(MDA)和超氧化物歧化酶(SOD)的含量。结果模型组、假手术组和低、高2个剂量实验组的筑巢实验评分分别为(2.67±0.33),(4.22±0.28)(3.89±0.42)和(4.22±0.15)分;这4组的MDA含量分别为(2.56±0.17),(1.13±0.10),(1.61±0.12)和(1.91±0.14)μmol·mg^(-1)。模型组的上述指标与假手术组比较,差异均有统计学意义(均P<0.01);低、高2个剂量实验组的上述指标与模型组比较,差异均有统计学意义(均P<0.05)。结论葛根素可能通过抗脑内氧化应激发挥改善缺血性脑卒中小鼠自理能力和空间学习记忆能力的神经保护作用。 Objective To observe the neuroprotection of puerarin against ischemic brain injury in mice by antioxidant stress.Methods According to the weight of mice,mice were randomly divided into 4 groups:sham group,model group,experimental-L group and experimental-H group,with 15 mice in each group.Before modeling,the mice in the experimental-L and experimental-H groups were intraperitoneally injected with puerarin(50,100 mg·kg^(-1))every day,for 4 d.Established the ischemic brain injury model by bilateral common carotid artery oeclusion 30 minutes reperfusion on the fifth day.One day after the operation,the mice in the experimental-L and experimental-H groups were given the drug continuously for 8 d.Mice in the sham and model groups were intraperitoneally injected with normal saline.Nest building activity experiment was used to evaluate the self-care ability of mice in daily activities.The Y maze experiment was used to evaluate the spatial working memory of mice.Malonaldehyde(MDA)and superoxide dismutase(SOD)contents was detected by kits.Results The nest building activity experiment scores in model group,sham group,experimental-L group and experimental-H group were(2.67±0.33),(4.22±0.28),(3.89±0.42)and(4.22±0.15)point;the content of MDA in the 4 groups were(2.56±0.17),(1.13±0.10),(1.61±0.12)and(1.91±0.14).Compared between model group and sham group,the difference of indicators were significant(all P<0.01);compared among experimental-L group,experimental-H group and model group the difference of indicators were significant(all P<0.05).Conclusion Puerarin may play a neuroprotective role in improving the self-care ability and spatial working memory ability of mice that are ischemic stroke by resisting oxidative stress.
