蒲公英多糖对急性肾损伤大鼠的保护作用研究 被引量:11

Protective effect of taraxacum polysaccharide on acute kidney injury rats

摘要 目的观察蒲公英多糖对急性肾损伤(AKI)大鼠的保护作用。方法通过腹腔注射140 mg·kg^(-1)庆大霉素构建AKI模型,并随机分为模型组和低、高剂量实验组,每组18只;另选取18只雄性Wistar大鼠作为正常组。造模结束24 h后,低、高剂量实验组按0.01 mL·g^(-1)·d^(-1)的给药剂量分别灌胃给予50和400 mg·kg^(-1)蒲公英多糖;正常组和模型组均灌胃给予等量0.9%NaCl。4组大鼠每天给药1次,连续给药7 d。用WST-8法检测大鼠肾组织中超氧化物歧化酶(SOD)的活性,用实时荧光定量聚合酶链反应法检测各组大鼠肾组织中SOD-1和白细胞介素-10(IL^(-1)0)mRNA的表达水平。结果给药7 d后,低、高剂量实验组和模型组、正常组的SOD分别为(44.19±6.48),(65.49±7.31),(35.84±6.29)和(79.32±7.57)U·mg^(-1),SOD-1 mRNA分别为0.23±0.11,0.45±0.09,0.16±0.07和0.48±0.11,IL^(-1)0 mRNA分别为0.21±0.07,0.41±0.10,0.17±0.08和0.47±0.11,低、高剂量实验组的上述指标与模型组比较,差异均有统计学意义(均P<0.05)。结论蒲公英多糖对急性肾损伤大鼠有保护作用,其机制可能与减轻炎症及氧化应激反应有关。 Objective To investigate the protective effect and mechanism of dandelion polysaccharide on acute kidney injury rats.Methods The rats were injected intraperitoneally with 140 mg·kg^(-1)gentamicin to create a model,and all of them were divided into model group,experimental-L group and experimental-H group,with 18 cases per group.Then 18 Wistar rats were severed as normal group.The experimental-L group and experimental-H group were given 50 and 400 mg·kg^(-1)dandelion polysaccharide,respectively,normal group and model group were given equal volume of saline.4 groups of rats were administered once a day for 7 days.The activity of superoxide dismutase(SOD)in kidney tissue of rats in each group was detected by WST-8.The levels of SOD-1 mRNA and interleukin-10(IL-10)mRNA in kidney tissue of rats in each group were detected by real-time polymerase chain reaction,the protein levels of SOD-1 and IL-10 in kidney tissue of rats in each group were detected by Western blot.Results After administration of 7 days,the main indexes of experimental-L,experimental-H,model and normal groups were compared:the levels of SOD were(44.19±6.48),(65.49±7.31),(35.84±6.29)and(79.32±7.57)U·mg^(-1),the expression levels of SOD-1 mRNA were 0.23±0.11,0.45±0.09,0.16±0.07 and 0.48±0.11,the expression levels of IL-10 mRNA were 0.21±0.07,0.41±0.10,0.17±0.08 and 0.47±0.11,there were statistically significant differences between experimental-L,-H groups and model group(all P<0.05).Conclusion Taraxacum polysaccharide could protect the kidney of rats with acute renal injury and its mechanism may be related to alleviating inflammatory reaction and oxidative stress.
