摘要
目的考察隐丹参酮制剂对肝性脑病大鼠的保护作用。方法SD大鼠随机分为正常组、模型组、谷氨酸片(360 mg/kg)组(阳性对照)、隐丹参酮高剂量(180 mg/kg)组、隐丹参酮中剂量(60 mg/kg)组、隐丹参酮低剂量(20mg/kg)组,每组10只。通过四氯化碳(CCl4)诱导慢性肝损伤伴高血氨模型、门腔静脉侧-侧分流术致大鼠肝性脑病模型,观察隐丹参酮对血清中丙氨酸氨基转移酶(ALT)、天门冬氨酸氨基转移酶(AST)、碱性磷酸酶(ALP)、总胆红素(TBIL)、总蛋白(TP)、白蛋白(ALB)水平影响,以及对血氨、脑组织氨含量、肝脏组织病理学的影响。结果在CCl4致大鼠慢性肝损伤伴高血氨模型中,与模型组比较,隐丹参酮中剂量组可显著降低血氨水平(P<0.05),隐丹参酮高剂量组能明显降低血氨及脑组织氨水平(P<0.05),且能改善大鼠肝功能,降低血清中AST和ALP水平,差异均有统计学意义(P<0.05);隐丹参酮中、高剂量组可改善肝组织变性、减轻肝坏死。在门腔静脉侧-侧分流术致大鼠肝性脑病模型中,与模型组比较,隐丹参酮中、高剂量组及谷氨酸片组血氨和脑组织氨水平明显降低(P<0.05),且隐丹参酮高剂量组能改善肝脏功能,降低模型大鼠血清中ALT、ALP、TBIL水平,差异均有统计学意义(P<0.05),且隐丹参酮中、高剂量组能减轻肝组织变性、炎症、坏死。结论隐丹参酮制剂对肝性脑病大鼠具有一定保护作用。
Objective To investigate the protective effect of cryptotanshinone preparations on rats with hepatic encephalopathy.Methods SD rats were randomly divided into normal group,model group,positive glutamate tablet(360 mg/kg)group,cryptotanshinone high-dose(180 mg/kg)group,cryptotanshinone medium-dose(60 mg/kg)group,cryptotanshinone low-dose(20 mg/kg)group,with 10 rats in each group.Hepatic encephalopathy animal models were induced by carbon tetrachloride and side-to-side portal-caval shunt surgery.The effects of cryptotanshinone were measured by detecting the content of alanine aminotransferase(ALT),aspartate aminotransferase(AST),alkaline phosphatase(ALP),total bilirubin(TBIL),total protein(TP),albumin(ALB)in the serum and brain tissue ammonia content.HE staining of liver tissue and histopathological examination were measured.Results In chronic liver injury with high blood ammonia rats caused by CCl4,the high and middle doses of cryptotanshinone could significantly reduce blood ammonia and brain tissue ammonia levels(P<0.05).The high dose of cryptotanshinone could improve liver function and reduce serum AST and ALP contents in in rats(P<0.05).In hepatic encephalopathy rats caused by side-to-side portal-caval shunt surgery,cryptotanshinone could reduce blood ammonia and brain tissue ammonia levels(P<0.05);cryptotanshinone could also improve nerve reflex,reduce animal mortality,improve liver function pathological damage.Conclusion The cryptotanshinone played an important role in protecting rats from hepatic encephalopathy.
