基于UPLC-Q-TOF/MS的复方鱼腥草合剂大鼠血清药物化学研究 被引量:7

Identification of absorbed components in rat plasma after oral administration of Yuxingcao Mixture by UPLC-Q-TOF/MS

摘要 目的采用超高效液相色谱-四极杆-飞行时间质谱联用技术(UPLC-Q-TOF/MS)对复方鱼腥草合剂口服吸收入血成分进行辨识研究,初步阐明其可能的药效物质基础。方法采用Acquity BEH C_(18)(100 mm×2.1 mm,1.7μm)色谱柱,流动相为0.1%甲酸水溶液(A)-乙腈(B),梯度洗脱,体积流量0.2 mL/min,柱温35℃,进样量5μL。采用电喷雾离子源,正、负离子模式扫描,毛细管电压负离子模式2 kV。离子源温度110℃,锥孔电压30 V,锥孔气体积流量50 L/h,氮气脱气温度350℃,脱气体积流量800 L/h,扫描范围m/z 50~2000。结果通过对复方鱼腥草合剂样品和空白组大鼠血浆色谱图及质谱碎片信息进行比对分析,鉴定出24个化学成分,包括15个原型成分和9个代谢产物。结论血浆中鉴定出的原型成分和代谢产物,可能是复方鱼腥草合剂潜在的活性成分,为其质量标志物研究提供了依据。 Objective To analyze and identify the chemical components in plasma from Yuxingcao Mixture by UPLC-Q-TOF/MS.Method Acquity BEH C_(18)(100 mm×2.1 mm,1.7μm)was used for isolation and identification of chemical components in Yuxingcao Mixture with a mobile phase of acetonitrile-water containing 0.1%formic acid for gradient elution and the flow rate of 0.2 mL/min.The column temperature was 35℃,and the sample size was 5μL.Electrospray ion(ESI)source were applied for the qualitative analysis under the positive and negative ion mode.The capillary voltage under negative ion mode was 2 kV,ion source temperature 110℃,cone hole voltage 30 V,cone hole gas volume flow rate 50 L/h,nitrogen degas temperature 350℃,degas volume flow rate 800 L/h,and scan range was m/z 50—2000.Result Based on UPLC-Q-TOF/MS analysis,a total of 24 compounds(including 15 prototypes and nine metabolites)were identified by comparing with ion chromatograms and pyrolysis information of mass fragmentation of the blank biological samples and Yuxingcao Mixture samples.Conclusion The prototype components and metabolites identified in the plasma may be the potential active components of Yuxingcao Mixture,which provides a foundation for the establishment of quality markers.
